Včelí choroby, zvláště bakteriálního původu, jsou v současnosti velkým problémem v chovu včel, protože způsobují rozsáhlé úhyny včelstev s významnými ekonomickými a ekologickými škodami. Zkoumání imunity včel může přispět k nalezení podstaty odolnosti včelstev a snížit jejich úhyny. Tato bakalářská práce je zaměřena na antimikrobiální peptidy včel jako součásti humorální imunity včel. V teoretické části práce byla zpracována literární rešerše o výskytu, složení, funkci a metodách analýzy antimikrobiálních peptidů včel. Dle zjištěných údajů z literatury byly navrženy a optimalizovány metody elektroforetického rozdělení malých peptidů s následnou detekcí v gelech a na blotovacích membránách. Nejlepšího výsledku bylo dosaženo při použití 16% polyakrylamidového gelu s přídavkem 6M močoviny. Tato metoda je využitelná pro analýzu peptidů v purifikované hemolymfě nebo purifikovaném extraktu včel. Metodou HPLC na reverzní fázi byl zjištěn retenční čas standardu antimikrobiálního peptidu apidaecinu na koloně Jupiter Proteo 90{\AA}, retenční čas byl poté ověřen pomocí metody MALDI-TOF. S použitím standardu apidaecinu byla dále optimalizována metoda nanoLC-ESI-MS/MS a použita pro detekci apidaecinu ve vzorcích včel.Honey bee disseases, especially bacterial, are actually a big problem in beekeeping. They cause the decrease of great number of honey bee colonies, with important economic and environmental consequences. Studies of honey bees` immunity can contribute to the understanding of bee resistance and would help to decrease the loss of honey bee colony. This bachelor thesis is focused on antimicrobial peptides. Antimicrobial peptides represent a part of the humoral innate immunity of insects. The theoretical section consists of the literature research about the incidence, structure, function and methods of analysis of antimicrobial peptides. According published literature, methods of electrophoretic separation and detection of small peptides in gels and on blotting membranes were designed and optimised. The use of 16% polyacrylamide gels with 6M urea gave the best results of peptide separation. This method is usefull for the analysis of the peptides in purified bee haemolymph or in honey bee extracts. The retention time of the antimicrobial peptide apidaecin was determined with the HPLC method on reverse phase on the collumn Jupiter Proteo 90{\AA} and verified by MALDI-TOF analysis. A nanoLC-ESI-MS/MS analysis was used for detection of apidaecin in samples using peptide standard to optimalise this method.
