Růst a vývoj rostlin je regulován rostlinnými hormony, které řídí mnoho fyziologických procesů. Auxiny, jedny z fytohormonů, hrají významnou roli při prodlužování kořene, jelikož indukují dělení buněk v kořenovém meristému a také se podílí na vývoji embrya. Již dříve byla publikována odlišná distribuce fytohormonů v jednotlivých orgánech, ale i na úrovni pletiva. Jejich rovnovážný stav, homeostáza, je ovlivněna jak biosyntézou, metabolismem tak transportem. Fytohormony mohou být transportovány symplastickou (buněčnou) i apoplastickou (mezibuněčnou) dráhou. Předpokládá se, že v těchto částech jsou koncentrace fytohormonů rozdílné. Cílem této práce je studium vlivu auxinových inhibitorů biosyntetických drah či transportu narušující homeostázu auxinů v Arabidopsis thaliana, kterou díky těmto látkám lze studovat. V praktické části byla provedena optimalizace izolace apoplastu a symplastu z kořenů nebo prýtů a podmínek inkubace A. thaliana se zvolenými syntetickými inhibitory. Dále byla provedena purifikace fytohormonů za využití metody extrakce pevnou fází (SPE) s jejich následným stanovením pomocí metody kapalinové chromatografie spojené s tandemovou hmotnostní spektrometrií (LC-MS/MS). Výsledky ukázaly rozdílnou distribuci auxinů a cytokininů mezi symplastem a apoplastem, fenotypové změny a ovlivněné fytohormonální profily po ošetření rostlin auxinovými inhibitory.Plant growth and development is regulated by plant hormones that control many physiological processes. Auxins, one of the phytohormones, play an important role in root elongation, throught the induction of cell division in the root meristem, and also participate in embryo development. Different distribution of phytohormones in individual organs, but also at the tissue level, has been previously published. Their equilibrium state, homeostasis, is affected by biosynthesis, metabolism and transport. Phytohormones can be transported by both the symplastic (cellular) and apoplastic (intercellular) pathways. It is assumed that the concentrations of phytohormones are different in these compartments. The aim of this work is to study the effect of auxin inhibitors of biosynthetic pathways or transport that disrupting auxin homeostasis in Arabidopsis thaliana that can be studied applying these substances. In the practical part, extraction, the isolation of apoplast and symplast from roots or shoots and incubation conditions of A. thaliana with selected synthetic inhibitors was performed. Furthermore, purification of phytohormones was done using the solid phase extraction (SPE) method, followed by liquid chromatography with tandem mass spectrometry (LC- MS/MS). The results showed different distribution of auxins and cytokinins between symplast and apoplast, phenotypic changes and affected phytohormonal profiles after treatment of plants with auxin inhibitors.
