Tato bakalářská práce se zabývá studiem patogenní variability padlí na salátu (Golovinomyces bolayi). V první části této práce byl vypracován podrobný popis rodu Lactuca, padlí Golovinomyces bolayi a přehled problematiky patogenní variability. Praktická část byla založena na testování izolátů G. bolayi na diferenciačním souboru 13 genotypů Lactuca spp. Testování probíhalo v letech 2019 a 2020 s izoláty posbíranými ze střední a jižní Moravy. Testování probíhalo v laboratorních podmínkách metodou listových disků pocházejících z rostlin předpěstovaných ve skleníku. Hlavním cílem bylo posouzení virulence v přirozených populacích patogena a určení specifity v interakcích Lactuca spp. - G. bolayi. Studie potvrdily, že interakce v tomto patosystému jsou specifické a závislé na genotypu napadené rostliny. Největší procento náchylnosti vykazoval genotyp L. serriola (LSE/57/15) a odolnost vykazovaly nejvíce kultivary L. sativa Colorado a L. virosa (LVIR/50). Celkově byl počet rezistentních reakcí nízký. Tyto výsledky byly dále porovnány s patogenní variabilitou padlí tykvovitých, kdy bylo zjištěno, že u všech studovaných patosystémů je patogenní variabilita vysoká a rezistence rasově specifická a mění se v průběhu let.This bachelor thesis is focused on study of pathogenic variability of powdery mildew on lettuce (Golovinomyces bolayi). The theoretical part of this thesis includes a detailed description of Lactuca spp., powdery mildew Golovinomces bolayi and pathogenic variability in general. The practical part was based on testing of isolates of G. bolayi on a differential set of 13 genotypes of Lactuca spp. Experiments were realized in 2019 and 2020 with powdery mildew isolates collected from central and south Moravia. Testing was performed under laboratory conditions with leaf discs method on plants grown in a greenhouse. The main objective was to assess virulence in natural populations of the pathogen and to determine specificity of the interactions of Lactuca spp. - G. bolayi. Studies have confirmed that interactions in this pathosystem are specific and dependent on the genotype of the infected plant. The genotype L. serriola (LSE/57/15) showed the highest percentage of susceptibility and cultivars L. sativa Colorado and L. virosa (LVIR/50) showed the greatest resistance. Overall, the number of resistant responses was low. These results were compared with the pathogenic variability of cucurbit powdery mildew, where was found that the pathogenic variability is high and resistance is race-specific for all studied pathosystems and changes over the years.
