Komplex ribosomálních kanálů [rukopis] / Monika Smékalová. -- 2009. -- 91 : il., tab. -- Ved. práce Michal Otyepka. -- Abstract: Tato práce shrnuje základní i detailní informace o komplexu ribozomálních kanálů nutných pro translokaci nascentního polypeptidu. Uvedeny jsou historické poznámky, současné poznatky i dosud neobjasněné skutečnosti. Práce vymezuje místo ribozomálních kanálů v genové expresi, popisuje jejich funkce i strukturu. Otázka translokace polypeptidů je spojena se strukturou ribozomu. Jelikož ribozom se liší v eukaryotických a prokaryotických organismech, nalezne zde čtenář i průběžné srovnávání těchto organel. Podstatnou část práce věnuji koordinované koexpresi proteinů, jako nástroje pro genovou terapii. Každé téma provází nejasnosti, ke kterým podávám možná vysvětlení a možnosti řešení.. -- Abstract: This work summarizes both basic and detailed information about the ribosomal channel complex, which are necessary for the translocation of a nescent polypeptide. There are also stated historical notes, recent knowledge as well as some realia which have not yet been sufficiently clarified. This thesis specifies the role of ribosomal channels in the gene expression and describes their functions and structure. The question of the polypeptide translocation is connected with the structure of the ribosome. As ribosomes differ in eukaryotic and prokaryotic organisms, the reader will find here also paraller comparison of these organelles. Substational part of my work is devoted to the co-ordinated co-expression of proteins as an tool for the gene therapy. Every subjects are accompanied with unclarities to which I give possible explanations and suggest possible solutions.
