Karcinom prostaty je onemocnění odvíjející se od chybného fungování androgenového receptoru (AR). Rakovinu prostaty lze podle závislosti na AR rozdělit na dvě stádia: prvním stádiem je tzv. na androgenech-závislá rakovina prostaty, která ke svému růstu a progresi vyžaduje aktivní AR a standardní léčba spočívá v chirurgickém nebo medikamentózním odstranění androgenů. Druhým stádiem je rakovina prostaty na androgenech-nezávislá, která se často vyvíjí z prvního stádia a charakteristickým rysem tohoto bujení jsou růst a progrese nádorových buněk nezávisle na AR. Předpokládali jsme, že gen kódující AR může být u na androgenech-nezávislého stádia nádorů prostaty transkripčně neaktivní a příčinou jeho umlčování - silencing, mohou být některé z epigenetických modifikací (především metylace DNA). Na androgenech-nezávislé stádium je známkou agresivní formy nádoru, který nereaguje na standardní léčbu. Proto se v posledních letech vyvíjejí a zkouší nové postupy a léčiva. Předkládaná práce posuzuje účinky inhibitoru metylací DNA (5-aza-2´-deoxycytidin, 5-AzA-CdR), a nebo jeho kombinaci s inhibitorem histonových deacetyláz (butyrát sodný, NaB) na metylaci DNA genu AR a tumor-supresorového genu ZNF185. Jak vyplývá z literatury, tyto geny bývají u na androgenech-nezávislých karcinomů prostaty transkripčně neaktivní. Inhibitory DNA metylací a inhibitory histonových deacetyláz jsme aplikovali v různých koncentracích po dobu 48 hod a 6 dní buňkám dvou prostatických nádorových liniích: PC-3 a DU-145. Výsledné změny v metylaci DNA byly posuzovány na DNA i cDNA úrovni a byla prokázána demetylace DNA genu AR u obou buněčných linií.Prostate cancer is one of the most frequently diagnosed cancer diesease, which is caused by disfunction of androgen receptor (AR). The AR mediates the effects of androgens, which are important in the development and maintenance of prostate cancer progression. In the initial stages, prostate cancer cells depend on androgens for survival, and the main form of treatment is androgen-withdrawal including surgical and/or androgen ablation therapy. However, in most cases the disease becomes progressive. The second stage of the disease is referred to as an androgen-independent prostate cancer. The main characteristic trait of this cancer is a growth and a progress of the cancer cells independently of AR. We assumed that in androgen-independent prostate cancer, the AR gene is transcriptional inactive and frequently silenced by epigenetic modifications (mainly by DNA methylation). This advanced stage of prostate cancer is unresponsive to standard therapy. Recently there have been developed new medical treatments and drugs for treatment of this disease. In the diploma thesis we examine the effect of inhibitor of DNA methylation (5-aza-2´-deoxycytidin, 5-AzA-CdR) and/or combine effect of this inhibitor with inhibitor of histon deacetylases (sodium butyrate, NaB) on methylation levels of the AR gene and/or ZNF185, potential tumor-suppressor gene. It is known that these genes are transcriptional inactive in androgen-independent prostate cancers and restoring of their expression could be possible way to prostate cancers arrest. The cells of two androgen-independent prostate cancer lines (DU-145 and PC-3) were treated with the inhibitors in different concentrations for 48 hours and 6 days. We analysed the demethylation effect of these inhibitors on both DNA and cDNA levels and we demonstrated DNA demethylation in the AR gene in both cell lines.
