Diplomová práce je věnována charakterizaci mutanta Arabidopsis thaliana, který má z funkce vyřazen gen kódující protein z NPH3 (non-phototrophic hypocotyl 3) rodiny. Tento gen je kandidátem pro polozakrslé vysokoplodící variety sóji luštinaté (Glycine max). V teoretické části práce jsou popsány nejen tyto specifické variety, ale také elitní linie sóji a divoký předek Glycine soja. Dále je rešerše zaměřena na rostliny a jejich vnímání světla, fotoreceptory a mechanismy reakce na stinné podmínky. Jedna z kapitol je věnována funkci NPH3 proteinu. V praktické části bylo nejprve ověřeno, zda semena loss-of-function nph3 mutanta A. thaliana (SALK_023877C), která byla získána z NASC (Nottinghamský inventář semen Arabidopsis), mají ve svém genomu T-DNA inserci. K tomu byly využity bioinformatické nástroje a molekulárně biologické metody. Následně byla zkoumána odpověď rostlin divokého typu, nph3 mutanta a dalších loss-of function mutantů jednotlivých fotoreceptorů při různých světelných podmínkách. Byly provedeny dva experimenty, kdy byla sledována odpověď rostlin při ošetření modrým světlem. Při prvním z nich byl sledován růst Arabidopsis při různé intenzitě modrého světla. Druhý experiment se zaměřoval na test fototropismu rostlin. Při obou experimentech se nph3 mutant choval stejně, jako divoký typ. Těmito experimenty nebyl potvrzen kandidátní gen pro polozakrslé vysokoplodící variety sóji luštinaté.This master's thesis is devoted to the characterization an Arabidopsis thaliana mutant with loss-of-function gene, that codes a protein of NPH3 (non-phototrophic hypocotyl 3) family. This gene is a candidate of semidwarf high-yielding varieties of soybean (Glycine max). In the theoretical part are described these unusual varieties, elite soybean lines and the wild ancestor Glycine soja. The research is then focused on plant light perception, photoreceptors, and mechanisms, that enable plants to react to shady environment. One chapter is devoted to the function of the nph3 gene. In experimental part is at fist verified, that the seeds of loss-of-function nph3 mutant of A. thaliana (SALK_0235877C), which were obtained from NASC (Nottingham Arabidopsis Stock Centre), have a T-DNA insertion in their genotype. Bioinformatical tools and methods of molecular biology were used to do so. Reaction of wild type plants, the nph3 mutant and another loss-of-function mutants of different photoreceptors were examined in different light conditions. Two light experiments were performed. The first one was blue light treatment, when Arabidopsis plants were grown in various intensities of blue light. The second experiment was about testing phototrophic response of the plants. Both experiments showed no significant difference between behavior of the wild type plants and the nph3 mutant. These experiments did not confirm the candidate gene of the semidwarf high-yielding varieties of soybean.
