Cílem této bakalářské práce bylo objasnit vliv proteinu PsbR na organizaci fotosyntetického aparátu v tylakoidní membráně u rostlin bez světlosběrných antén Lhcb3 a Lhcb6. U proteinu PsbR je známo, že je podstatný pro stabilitu a správnou funkci fotosyntézy. V nejnovějších prací byla navržena hypotéza, že se protein PsbR může podílet na interakci mezi sousedními komplexy fotosystému II skrze jejich stromální stranu, a že může být důležitý pro tvorbu granálních tylakoidů. Dále bylo zjištěno, že se PsbR preferenčně vyskytuje v komplexech fotosystému II s menším množstvím světlosběrných antén, což napovídá i situaci např. v mutantech Arabidopsis thaliana bez proteinů Lhcb3 a Lhcb6. V rámci bakalářské práce byl pomocí křížení linií kolhcb6 a kopsbR připraven dvojitý mutant a pomocí kolhcb3kolhcb6 a HZ (lhcb3lhcb6psbR) trojitý mutant. U mutantů byla charakterizována fotosyntetická funkce pomocí metody chlorofylové fluorescence a následně byla studována organizace tylakoidních membrán a komplexů fotosystému II pomocí transmisní elektronové mikroskopie a nativní elektroforézy. Prezentované výsledky se přiklání k hypotéze, že protein PsbR může mít vliv na interakce mezi fotosystémy II a přispívat tak ke strukturní organizaci tylakoidní membrány.The aim of this bachelor thesis was to elucidate the influence of PsbR protein on the organization of the photosynthetic apparatus in the thylakoid membrane in plants without light-harvesting antennas Lhcb3 and Lhcb6. The PsbR is known to be essential for the stability and correct functioning of photosynthesis. Recent work suggests that the PsbR may be involved in the interaction between neighboring photosystem II complexes through their stromal side, and that it may be important for the formation of granular thylakoids. Furthemore, it was founded that PsbR preferentially occurs in complexes of photosystem II with smaller number of light-harvesting antennas, which suggests the situation, for example, in Arabidopsis thaliana mutants without Lhcb3 and Lhcb6 proteins. In the frame of the bachelor´s thesis, a double mutant was prepared by crossing the kolhcb6 and kopsbR lines and a triple mutant using kolhcb3kolhcb6 and HZ (lhcb3lhcb6psbR). In mutants, photosynthetic function was characterized by chlorophyll fluorescence method and than the organization of thylakoid membranes and photosystem II complexes was studied using transmission electron microscopy and native electrophoresis. The presented results support the hypothesis that the PsbR may affect the interaction between photosystem II and thus to the structural organization of the thylakoid membrane.
