Rastová reštrikcia u dieťaťa môže byť spôsobená exogénnymi vplyvmi, môže byť jedným z prvých príznakov iného systémového ochorenia, ale tiež môže byť zapríčinená genetickou poruchou. Jednou z možností na stanovenie prítomnosti genetickej poruchy je sekvenovanie génov ovplyvňujúcich vzrast dieťaťa. Medzi takéto gény patria NPR2, ACAN, IGF1, IGF1R, FGFR3, COL2A1, GHR, STAT5B, IGFALS, SHOX. Prvým z cieľov tejto práce bolo vyhodnotiť dáta získané masívne paralelným sekvenovaním regulačných oblastí génu SHOX, ktorých význam pre rast človeka doposiaľ nie je dostatočne preskúmaný. Ďalej bolo cieľom tejto práce vykonanie segregačnej štúdie vybraných génov súvisiacich s idiopaticky nízkym vzrastom v troch rodinách. Varianty odhalené touto štúdiou boli následne podrobené case control štúdií v súbore 100 vzoriek DNA od pacientov nevykazujúcich fenotyp nízkeho vzrastu. Bioinformatická analýza sekvenačných dát regulátorov génu SHOX odhalila 3 doposiaľ nepopísané CNV varianty (v CNE-5 a CNE7) a 4 varianty s frekvenciou výskytu nižšou ako 0,5 % (v CNE3, CNE5, CNE8, CNE9). Tieto nálezy sú vhodným cieľom pre ďalšie klinické štúdie. Segregačná štúdia odhalila 3 suspektné varianty, a to v génoch FGFR3 (NM_000142.4:c.2005C>G), COL2A1 (NM_001844.5:c.1636G>A) a GHR (NM_000163.5:c.440-1G>A). Case control štúdia nepotvrdila prítomnosť týchto variantov v negatívnych kontrolách, a tak ide pravdepodobne o patologické varianty, ktoré zapríčiňujú fenotyp nízkeho vzrastu.Growth restriction in a child can be caused by exogenous influences, it can be one of the first symptoms of another systemic disease, but it can also be caused by a genetic disorder. One way to determine the presence of a genetic disorder is to sequence genes that affect a child's growth. Such genes include NPR2, ACAN, IGF1, IGF1R, FGFR3, COL2A1, GHR, STAT5B, IGFALS, SHOX. The first aim of this work was to evaluate the data obtained by massively parallel sequencing of the regulatory regions of the SHOX gene. Significance of these regions for human growth has not yet been sufficiently investigated. Furthermore, the aim of this work was to perform a segregation study of selected genes related to idiopatic short stature in three families. The variants revealed by this study were subjected to case control study in a set of 100 DNA samples from patients without the phenotype of short stature. Bioinformatics analysis of SHOX gene regulator sequencing data revealed 3 unknown CNV variants (in CNE-5 and CNE7) and 4 variants with an incidence of less than 0,5% (in CNE3, CNE5, CNE8, CNE9). These findings are a suitable target for further clinical trials. The segregation study revealed 3 suspect variants, namely in the genes FGFR3 (NM_000142.4: c.2005C> G), COL2A1 (NM_001844.5: c.1636G> A) and GHR (NM_000163.5: c.440-1G> A ). The case control study did not confirm the presence of these variants in the negative controls, and thus they are probably pathological variants that cause a low-growth phenotype.
