Rod máty (Mentha L.) je široce využíván v zemědělství, potravinářském průmyslu a medicíně. Důležitým parametrem pro zhodnocení kvality rostlin máty je obsah sekundárních metabolitů. Teoretická část diplomové práce se zabývá hydroponickým pěstováním máty. Práce je zaměřena na abiotický stres rostlin a vliv vybraných stresorů na metabolismus dusíku a na obsah sekundárních metabolitů. Práce byla provedena s cílem sledování vlivu abiotického stresu na složení tří vybraných genotypů máty a zhodnocení jejich kvality. V praktické části byla pomocí tří experimentů provedena optimalizace hydroponického pěstování rostlin máty. Rostliny byly pěstovány za kontrolovaných podmínek. Kontrolním roztokem bylo Hoaglandovo médium o ? síle (? HM). V průběhu pěstování byly rostliny stresovány sníženým i zvýšeným příjmem dusičnanů a solným stresem a byly sledovány změny v hladinách jednotlivých metabolitů. Stěžejním experimentem bylo semihydroponické pěstování ve fenotypizačním systému Plantscreen conveyor, kdy bylo prováděno měření fluorescence chlorofylu a RGB zobrazení rostlin. Pomocí ultra vysokoúčinné kapalinové chromatografie s hmotnostní detekcí (UHLPC-MS/MS) byla provedena analýza volných polyaminů, aminokyselin a fenolických sloučenin v rostlinném materiálu. Majoritně zastoupenou fenolickou sloučeninou byla ve všech vzorcích kyselina rozmarýnová. Identifikace a kvantifikace terpenoidů byla provedena s využitím plynové chromatografie s hmotnostní spektrometrií (GC-MS). Složení genotypů odpovídalo předchozím studiím. Majoritně zastoupeným terpenoidem M. arvensis byl karvon. Piperitenon oxid byl dominujícím terpenoidem genotypů M. piperita a M. spicata. U jednotlivých genotypů máty byla zaznamenána odlišná odezva na působení abiotického stresu. Z hlediska morfologie rostlin bylo nejvhodnějším médiem pro pěstování M. arvensis i M. piperita kontrolní ? HM, pro pěstování M. spicata to byl roztok se sníženou koncentrací dusičnanů. Jako nepříznivý pro pěstování genotypu M. arvensis se s ohledem na změny v hladinách metabolitů jevil stres příjmem dusičnanů. Signifikantní rozdíly mezi jednotlivými genotypy byly zaznamenány zejména v toleranci solného stresu. Jako nejtolerantnější se z hlediska složení aminokyselin, polyaminů a fenolických látek vůči solnému stresu jeví genotyp M. piperita, naopak nejméně tolerantním genotypem je M. spicata. Poznatky zjištěné v diplomové práci nasvědčují tomu, že studium podmínek pěstování jednotlivých genotypů máty je nezbytné pro zlepšování kvality rostlin.Plants of genus Mentha are widely used in agriculture, the food industry, and medicine. The content of secondary metabolites is an important parameter for the quality assessment of mint plants. The theoretical part of the diploma thesis deals with the hydroponic cultivation of mint. The thesis is focused on abiotic plant stress and the influence of selected abiotic stressors on nitrogen metabolism and the content of secondary metabolites. The thesis aimed to observe the influence of abiotic stress on the composition of three selected mentha genotypes and to evaluate their quality. In the experimental part of the thesis optimizing of hydroponic growth conditions of mint plants were performed using three consecutive experiments. Plants were cultivated under controlled conditions. The control solution was Hoagland half-strength medium (? HM). Plants were stressed during the cultivation using low and high nitrate intake and salt stress and changes in levels of metabolites were monitored. The main experiment was semi hydroponic cultivation in phenotyping system Plantscreen conveyor, where measurement of chlorophyll fluorescence and RGB imaging of plants was performed. Using ultra-high performance liquid chromatography with mass spectrometry (UHPLC-MS/MS) the analysis of free polyamines, amino acids, and phenolics in plant material was carried out. Rosmarinic acid was found to be the major phenolic compound in all genotypes. Identification and quantification of terpenoids were performed by gas chromatography-mass spectrometry (GC-MS). The composition of all mint genotypes corresponds with previous studies. The most abundant terpenoid of M. arvensis was carvone. Piperitenone oxide was the most dominant terpenoid of M. piperita and M. spicata genotypes. Different abiotic stress responses were observed among genotypes. Regarding plant morphology, the optimal solution for growing M.arvensis and M. piperita in hydroponics was a control solution (? HM), for M. spicata it was the solution with low nitrate concentration. Considering changes in metabolite levels, nitrate stress appeared to be unfavorable for the soilless growing of M. arvensis. Significant differences in salt stress tolerance between the selected genotypes were observed. According to amounts of free polyamines, amino acids, and phenolic compounds, M. piperita appears to be the most salt-tolerant. On the contrary, the least tolerant to salt stress seems to be genotype M. spicata. Findings of the diploma thesis suggest that the study of growth conditions in different mint genotypes is necessary for improving the quality of plants.
