Tato diplomová práce se zabývá syntézou a biologickou charakterizací peptidů s potenciálními antimikrobiálními vlastnostmi. V teoretické části jsou shrnuty základní vlastnosti antimikrobiálních peptidů, mechanismus jejich působení, funkce a zástupci. Dále je zde stručně popsáno získávání peptidů a zvláště pak metoda syntézy peptidů na pevné fázi. Experimentální část je zaměřena na přípravu látek, jejich charakterizaci dostupnými fyzikálně-chemickými metodami a biologickou analýzu. Při stanovení proteolytické stability bylo zjištěno, že peptidy obsahující v sekvenci D-aminokyseliny nejsou štěpeny enzymem trypsinem. Dále byla stanovena hodnota minimální inhibiční koncentrace peptidů pro bakterie Pseudomonas aeruginosa a Staphylococcus aureus. Pro stanovení cytotoxicity vůči savčím buňkám byly využity myší dermální fibroblasty. U některých peptidů byly prokázány koncentrace, které působí toxicky vůči bakteriálním buňkám, ale jsou zároveň netoxické vůči savčím. Další charakterizací bylo definování hemolytické aktivity s využitím lidské krve a v poslední řadě byly vybrané peptidy také testovány pro svou anti-biofilmovou aktivitu. Získané výsledky napovídají tomu, že struktura a vlastnosti peptidů hrají významnou roli při definování jejich antimikrobiální aktivity a že mohou tyto látky působit jako významná alternativa v boji proti bakteriálním onemocněním.The main topic of this diploma thesis is synthesis and biological characterization of potentially antimicrobial peptides. In the theoretical part, main features of antimicrobial peptides (AMPs) are introduced including mechanism of action, function and specimen of AMPs. Furthermore, methods of AMPs extraction are described with a focus on solid-phase peptic synthesis. Within materials and methods, detailed protocols of peptide synthesis and their biological examination are provided. Examination of proteolytic stability of the synthetized peptides indicated, that peptides with a D-amino acids in their sequence are not susceptible to cleavage by trypsin. Moreover, minimum inhibitory concentration values against Pseudomonas aeruginosa and Staphylococcus aureus bacteria were determined. Subsequently, mouse dermal fibroblasts were used for mammalian cytotoxicity measurement. For certain concentrations, some peptides displayed both effective bacterial killing and no mammalian toxicity. Hemolytic activity was assayed by using human blood and additionally anti-biofilm activity assessment was conducted as well. Findings of this research suggest that structure and properties are crucial in determination of peptide antimicrobial effects and support the idea of potential use of peptides in bacterial infection treatment.
