Bakalárska práca bola venovaná optimalizácii a následnej práci s protokolom pre stanovenie aktivity enzýmu MARK4, identifikovaného ako kľúčového pri hyperfosforylácii tau proteínu v oblastiach R1 a R3 MT-binding region. Pre uvedenú optimalizáciu bola použitá technika MALDI-TOF hmotnostná spektrometria, pomocou ktorej bola stanovená fosforylácia substrátu CHktid peptidu a zmena aktivity MARK4 vplyvom odlišných podmienok prostredia. Optimalizovaný protokol bol ďalej využitý pri testovaní inhibítorov, staurosporinu a MARK/PAR-1, hodnotených z hľadiska špecifity a vplyvu na uvedený enzým. Potvrdením funkčnosti metodiky nešpecifického staurosporinu a špecifického MARK/PAR-1 bolo možné pristúpiť k skríningu látok ako potenciálnych liečiv tvorby neurofibrilárnych klbiek, dôležitého ukazovateľa Alzheimerovej choroby. Okrem aplikovaného CHKtid peptidu bola funkčnosť metodiky overená i na peptidoch R1 a R3 tau proteínu.The bachelor thesis was dedicated to the optimization and determining the activity of the MARK4 enzyme, identified as key in the hyperphosphorylation of R1 and R3 peptides of the MT-binding region of tau protein. To determine the phosphorylation of the substrate CHktid peptide and the change of MARK4 activity due to different environmental conditions, MALDI-TOF mass spectrometry technique was used. The optimized protocol was further applied in testing of the inhibitors, staurosporine and MARK/PAR-1, with regards on specificity and effect on the enzyme. By confirming the functionality of the methodology of non-specific staurosporine and specific MARK/PAR-1, it was possible to proceed with the screening of substances as potential cure for the formation of neurofibrillary tangles, an important indicator of Alzheimer's disease. In addition to the applied CHK peptide, the functionality of the methodology was also verified on peptides R1 and R3 of tau protein.
