Cibule šalotka (Allium cepa var. aggregatum L.) je přirozeně dormantní rostlina významná pro svou chuť, obsahové látky a zdravotní benefity. Během dormance se mění její chemické složení, zvláště vzájemný poměr malých sacharidů a fruktanů vlivem fruktanexohydrolas. V šalotce je přítomna hlavně fruktan-1-exohydrolasa. V této práci byla zkoumána dynamika malých sacharidů, glukosidů kvercetinu a enzymová aktivita fruktan-1-exohydrolasy. Cílem bylo posoudit využití dynamiky obsahových látek a enzymové aktivity jako markerů ukončení dormance a začátku klíčení. Parametry byly sledovány v deseti kultivarech skladovaných při kontrolované teplotě během pěti měsíců. Obsah fruktosy a sacharosy byl stanoven metodou HPLC spojenou s odpařovacím detektorem rozptylu světla. V průběhu skladovaní měl obsah obou sacharidů mírně rostoucí trend. Šlo však o velmi proměnlivé parametry, závislé od množství faktorů, proto byl obsah sacharidů vyhodnocený jako nevhodný marker konce dormance. Glukosidy kvercetinu byli kvantifikované metodou HPLC s UV detekcí. Dynamika kvercetin-4'-O-glukosidu a kvercetin-3,4'-O-diglukosidu neměla v kultivarech shodný trend. Protože nesouvisí s metabolickými procesy při klíčení, byl obsah glukosidů kvercetinu vyhodnocený jako nevhodný marker. HPLC metoda stanovení aktivity fruktan-1-exohydrolasy byla v důsledku časté kontaminace a nízké spolehlivosti a reprodukovatelnosti vyhodnocená jako nevhodná.Shallot (Allium cepa var. aggregatum L.) is a naturally dormant plant, valued for its flavour, nutrition values and health benefits. Composition of shallot, especially the ratio between fructans and simple carbohydrates, changes during dormancy. This is due to the presence of fructan exohydrolases, primarily fructan 1-exohydrolase. Dynamic changes of simple carbohydrates and quercetin glucosides content, along with enzyme activity of fructan 1-exohydrolase, were the subject of this study. The aim of this study was to determine the applicability of mentioned parameters as dormancy release markers. These parameters were measured in ten cultivars during the storage period of five months in controlled temperature. The content of sucrose and fructose was determined using HPLC coupled with evaporative light scattering detector. The content of both saccharides increased slightly during storage. It was, however, a very variable parameter dependent on many factors, therefore it was rejected as a suitable dormancy marker. Quercetin glucosides were quantified using HPLC with UV detection. Dynamics of quercetin 3,4'-diglucoside and quercetin 4'-glucoside was observed to be variable amongst cultivars. In addition, content of these glucosides is not closely related to metabolic processes during sprouting. It was thus rejected as a suitable dormancy marker as well. Activity of fructan 1-exohydrolase was determined using HPLC. Because of frequent contamination, low reliability and low reproducibility, it was deemed an inappropriate method.
