Tato práce je zaměřena na stanovení obsahu cytokininů na buněčné úrovni u modelových rostlin Arabidopsis thaliana a Hordeum vulgare (ječmen). Rešerše v teoretické části se věnuje základní charakteristice cytokininů, jejich metabolismu a biologickým účinkům. Větší důraz je kladen na výskyt a metabolismus cytokininů uvnitř buňky. Praktická část obsahuje výsledky izolace protoplastů a vakuol z listů A. thaliana a ječmene, ve kterých byla následně stanovena hladina cytokininů. Pro izolaci protoplastů a vakuol, založené na enzymatickém odstranění buněčné stěny a následné separaci organel pomocí hustotního gradientu, byly použity 2 genotypy A. thaliana (wild type a mutantní forma wat1-1) a 3 genotypy ječmene (wild type, AtCKX1, HvCKX1). Ve vzorcích listů a získaných protoplastů a vakuol byla následně stanovena koncentrace 25 jednotlivých typů cytokininů pomocí UHPLC-MS/MS. Zjištěné množství cytokininů bylo porovnáno pomocí obsahu tonoplastové V-ATPasy a hodnoty byly následně vztaženy na extracelulární prostor, cytosol a vakuoly. Nejvyšší podíl cytokininů byl v případě Arabidopsis i ječmene mimo buňku, až 90 %, s majoritním zastoupením O- a N-glykosidů; přibližně 5 % cytokininů bylo poté obsaženo v cytosolu a ve vakuolách. V listech Arabidopsis byly nejpočetnější inaktivní formy cytokininů, převážně tZ7G, tZ9G and iP7G. Mutantní linie wat1-1 vykazovala oproti wild type formě výrazně vyšší obsah extracelulárního iP a iPR, obsah tZ glukosidů a iP glukosidů zůstal nezměněn. U obou linií A. thaliana byly porovnatelné hladiny derivátů DHZ a cZ, pouze v případě extracelulárního cZROG a cytosolárního cZR byl u wat1-1 detekován 5-násobný a 3-násobný nárůst. V případě ječmene byly pro stanovení hladiny cytokininů použity mladé listy. Celkový obsah cytokininů byl pro obě transgenní linie srovnatelný a v porovnání k wild type formě byl o trochu nižší. Nejvyšší rozdíl byl nalezen u extracelulárního tZ9G, kdy došlo u AtCKX1 i HvCKX1 k více než 50% poklesu. Linie AtCKX1 také vykázala 4x nižší obsah tZ mimo buňku a virtuálně žádný tZ uvnitř buňky. Oproti tomu obě transgenní linie obsahovaly dvakrát vyšší obsah extracelulárního iP a vakuolárního iPR. Přibližně 10% nárůst byl také detekován u cZOG mimo buňku. Metodu pro stanovení obsahu cytokininů popsanou v této práci je možné použít i pro analýzu dalších rostlinných hormonů možnost oddělit buněčné organely lze využít při studiu lokalizace, transportu a percepce fytohormonů v rostlinách.This thesis is focused on uncovering cytokinin distribution at cellular level for model plant Arabidopsis thaliana and Hordeum vulgare. The theoretical part deals with the general characterization of cytokinins, their metabolism and biological effects. The special attention is paid to the cellular localization of cytokinins and enzymes involved in their metabolism. The experimental part shows the results of the isolation of intact protoplasts and vacuoles obtained from leaves of A. thaliana and barley and the subsequent determination of cytokinin contents. Two genotypes of A. thaliana (wild type and wat1-1) and three genotypes of barley (wild type, AtCKX1, HvCKX1) were used for protoplasts and vacuoles isolations based on enzymatic removal of cell walls and separation of cell compartments at the Ficoll density gradient. Then, totally 25 unique cytokinin types were determined by UHPLC-MS/MS in the leaves and obtained protoplasts and vacuoles. Cytokinin quantities in extracellular space, cytosol and vacuoles were compared using tonoplast V-ATPase as a marker. The highest portion of cytokinins, almost 90 %, was located outside the cell in both plant species, with majority of O- and N-glycosides, while cytosol and vacuoles held approx. 5 % of total cytokinins each. In Arabidopsis matured leaves, the most abundant cytokinins were the inactive forms, especially tZ7G, tZ9G and iP7G. The wat1-1 mutant differed from the wild type by a significantly higher content of extracellular tZ and intracellular iP and iPR while the content of tZ and iP glucosides were unchanged. The levels of DHZ and cZ types were comparable for wild type and wat1-1, except the 5-fold and 3-fold increase of extracellular cZROG and cytosolic cZR in wat1-1. In barley young leaves, the total content of cytokinins was for AtCKX1 and HvCKX1 mutant lines comparable to each other and, in the same time, slightly lower than a wild type. The highest divergence was detected for the extracellular tZ9G with more than 50% decline in both transgenic lines. The AtCKX1 mutant showed about 4-fold decreased content of tZ in the extracellular space while intracellular tZ was virtually absent. Contrary to that, the 2-fold increase of extracellular iP and vacuolar iPR was detected in both mutants. Approx. 10% rise was also found for cZOG occurring in extracellular space of mutant lines. The developed experimental method can be applied to hormone analysis in general; the ability to separate some cellular compartments can help to understand to their localization, transport and perception in plants.
