Kritickým krokem v buněčném dělení je správné sestavení dělícího vřeténka, které napomáhá separaci chromozomů do dceřiných buněk. Jedním z regulačních proteinů je i TPX2, jenž ovlivňuje složení dělícího vřeténka a jeho délku u živočichů i rostlin. TPX2 protein navíc reguluje a aktivuje Aurora kinázy. Tyto mitotické kinázy jsou nedílnou součástí buněčného dělení, kde se fosforylací jiných proteinů účastní segregace chromozomů, nukleace mikrotubulů a cytokineze. Mutace TPX2 nebo Aurora kináz vede v buňce k narušení správného mechanismu dělení a může vést i k nádorovému bujení. Na rozdíl od živočišných buněk, které mají pouze jeden gen kódující protein TPX2, genom Arabidopsis thaliana kóduje 12 proteinů s podobnou strukturou, tzv. TPX2-like proteinů. Bylo zjištěno, že tyto proteiny obsahují různé kombinace funkčních domén přítomných v kanonickém TPX2, které ovlivňují jejich vlastnosti, a prochází alternativním sestřihem. Cílem první části této diplomové práce bylo umlčet kanonický TPX2 v rostlinách A. thaliana pomocí RNA interference. Tato metoda využívá specifické genové sekvence, které jsou zaklonovány do binárních vektorů a následně transformovány do rostlin s využitím bakterií Agrobacterium tumefaciens. Následně byla v rostlinách ověřována přítomnost RNAi konstruktů a exprese TPX2 genu pomocí polymerázové řetězové reakce. V posledním kroku byl ověřován vliv umlčení genu na tvorbu pylových zrn a semen. Expresi TPX2 genu se podařilo snížit ve 12 rostlinách. V druhé části byla zkoumána lokalizace vybraných TPX2-like proteinů v buňce metodou infiltrace listového mezofylu rostlin Nicotiana benthamiana fúzními konstrukty. Stejná metoda byla použita ke studiu možné regulace Aurora kinázy 1 TPX2-like proteiny. Byla sledována kolokalizace vybraných TPX2-like proteinů s Aurora kinázou 1 a proteinem CENH3.A critical step in the cell division is a correct assembly of the mitotic spindle which helps to separate chromosomes into daughter cells. One of the regulatory protein is TPX2 which influences the proper composition and lenght of the mitotic spindle in animals and plants. Moreover TPX2 protein regulates and activates Aurora kinase family members. These mitotic kinases are an integral part of the cell division where regulate chromosome segregation through phosphorylation of its targets including proteins involved in chromosome segregation, microtubule nucleation and cytokinesis. Mutations of TPX2 or Aurora kinases in the cell leads to disruption of proper mechanisms of the cell division and can lead to tumor growth. In contrast to animal cells which have only one gene encoding a TPX2 protein, Arabidopsis genome encodes 12 proteins with similar structure, TPX2-like proteins. TPX2-like proteins comprise different combinations of functional domains that are present in the canonical TPX2, therefore affecting its functional properties. The complexity is further influenced by alternative splicing The aim of the first part of this thesis was to silence canonical TPX2 in A. thaliana plants using RNA interference. This method use specific gene sequences that are cloned into binary vectors and transformed to plants using Agrobacterium tumefaciens mediated transformation. Subsequently, the presence of RNAi constructs and decreased TPX2 gene expression were checked using polymerase chain reaction. The effect of decreased TPX2 expression was further checked on the formation of pollen grains and seeds. The decreased expression of the TPX2 gene was verified altogether in 12 plants. In the second part of this thesis, the localization of selected TPX2-like proteins in Nicotiana benthamiana plants was studied. N. benthamiana leaf mesophyll cell were infiltrated with fusion constructs and localization was studied using confocal microscopy. Moreover, potential link between function of Aurora kinase-1 by TPX2 likes proteins was investigated by its colocalization with Aurora kinase 1 and CENH3.
