Tato práce je zaměřena na monitoring transkripční aktivity vybraných receptorů ovlivněné přítomností GO nanočástic. První část práce se věnuje teoretickému úvodu s charakteristikou testovaných receptorů (glukokortikoidního, androgenního a thyroidního) a dosud popsanými vlastnostemi GO. V praktické části je pomocí MTT testu hodnocena toxicita GO na sledované buněčné linie, která byla vyhodnocena jako žádná nebo zanedbatelná a pomocí GRA a PCR je sledována transkripční aktivita. V případě AZ-GR a PZ-TR buněčné linie se GO choval jako synergista v součinnosti s agonisty receptorů a s rostoucí koncentrací GO (při všech velikostech) rostla aktivace receptoru, která v obou případech přesáhla i 150 % aktivace samotným agonistou. V agonistickém módu dokázal GO aktivovat pouze TR a při velikosti částic > 500 nm při koncentraci 20 ?g/ml dosáhl téměř stejné aktivace jako T3 (pozitivní kontrola). Pouze u linie AIZ-AR se GO choval antagonisticky a jeho antagonistický účinek rostl s rostoucí koncentrací ve všech třech případech velikostí nanočástic. Pokles aktivity byl pro nejvyšší koncentrace až o 40 % oproti negativní kontrole (DHT). Výsledky metody PCR neposkytují dostatečné informace a vyžadují další studie. V agonistickém módu je u TR pozorovatelný trend snižující se exprese s rostoucí koncentrací GO.This work is focused on transcriptional activity of chosen receptors influenced by GO. In the first part of this work, there is a theoretical introduction of the characterization of tested receptors (glucocorticoid, androgen and thyroid receptors) and known characteristics of GO. Second part describes used methods and the results. The MTT test was used for measuring cytotoxic effects of GO on tested cell lines. No effect was observed. GRA and PCR methods were used for testing transcriptional activity. In the case of AZ-GR and PZ-TR cell lines, GO acted like synergist in combination with known agonists. With increasing concentration of GO (for all tested sizes), increased receptors activation was observed, which got higher than 150 % of agonist's activation for both receptors (GR, TR). GO alone activated TR and it almost reached the same value as T3 (positive control) for GO > 500 nm with concentration 20 ?g/ml. Antagonistic tendencies were recorded for AIZ-AR cell line only. There was significant decline of inducible luciferase activity with increasing concentrations of GO (for all tested sizes). In case of higher concentration, the inhibition was almost 40 % against negative control (DHT). Results of PCR method are inconclusive and require more studies. In agonistic mode for TR, there is a tendency of decreasing expression with increasing concentration of GO.
