Disertační práce "Screening chemických sloučenin in vitro" shrnuje výsledky z hodnocení analýzy cytotoxicity, modulace buněčného cyklu a antimikrobiálních aktivit souboru několika skupin látek ( nukleosidy, chelátory, deriváty cytosinu). Základní metodou studia mechanismů účinku protinádorových léčiv je buněčný cyklus. Modifikací této metody je hodnocení aktivity syntézy DNA a RNA a procentuální zastoupení buněk v mitóze. K pochopení mechanismu účinku je potřeba znalostí průběhu jednotlivých fází buněčného cyklu a jeho kontrolní mechanismy. Významnou roli zastávají ve výtěžnosti expresních analýz metody redukující komplexitu vzorku nejen na úrovni jedné buňky, ale i v populacích. Cílové buněčné populace můžeme na základě diferenciačních markerů vybrat pomocí metody průtokové cytometrie, konkrétně sortování buněk. Kromě protinádorových aktivit byly u látek sledována i jejich antimikrobiální aktivita jak u nejběžnějších bakteriálních kmenů a jejich resistetních variant, tak u specifických patogenů zpúsobujích závažné onemocnění, např. Mycobacterium. Výstupem této práce jsou: -Selekce derivátů s významným účinkem na průběh buněčného cyklu -selekce derivátů s významným antimikrobiálním účinkem -Validace identifikovaných kandidátních molekulDisertation thesis "Screening of chemical compounds in vitro" summarizes results from cytotoxicity, cell cycle modulation and antibacterial activity assays evaluation for set of compounds (nucleosides, chelators, cytosine derivatives). The primary method for the study of anticancer drugs´ mechanism of action represents cell cycle analysis. Evaluation of DNA and RNA synthesis activity, as well as percentage of cells in mitosis assesment, can be achived by modification of this method. For better understanding of mechanisms of action, it is necessary to understand the control mechanisms of cell cycle. Significant role for expression analysis represent methods reducing the complexity of sample not only for one cell, but in populations as well. Targeted cell populations can be sorted afterwards using flow cytometry sorting system. Antibacterial activities of tested compounds were evaluated as well. Activity against common bacterial strains (and their resistant equvalents) and specific pathogens, such as Mycobacterium, were obtained. Outcome of this thesis is: - selection of compounds with significant effect on cell cycle modulation - selection of compounds with significant antibacterial activity - validation of identified candidate molecules
