Ve své diplomové práci jsem hledala polymorfní mikrosatelitové lokusy u nesyta indomalajského (Mycteria leucocephala) metodou cross-species PCR amplifikace. Testovala jsem mikrosatelitové lokusy, které byly polymorfní u zástupců řádu trubkonosí (Procellariiformes). V teoretické části mé diplomové práce jsem uvedla základní charakteristiku řádu brodiví (Ciconiiformes), zejména čeledi čápovití (Ciconiidae). Zvláštní pozornost jsem věnovala rodu nesyt (Mycteria) a jednu kapitolu jsem věnovala popisu nesyta indomalajského. Dále jsem se zabývala charakteristikou řádu trubkonosí, uvedla jsem základní fakta o mikrosatelitech a teoreticky jsem popsala metodu cross-species PCR amplifikace. Závěrem jsem uvedla charakteristiku všech mikrosatelitových lokusů, které byly polymorfní u zástupců řádu trubkonosí. V praktické části mé práce jsem hledala polymorfní mikrosatelitové lokusy u nesyta indomalajského. Použila jsem metodu cross-species PCR amplifikace a testovala jsem 212 mikrosatelitových lokusů, z toho 207 lokusů pocházejících od druhů řádu trubkonosí, 4 mikrosatelitové lokusy pocházející od zástupců řádu dlouhokřídlí a 1 mikrosatelitový lokus pocházející od druhu z řádu pěvci. Všechny tyto lokusy byly polymorfní u zástupců řádu trubkonosí. U nesyta indomalajského jsem nalezla celkem 46 polymorfních mikrosatelitových lokusů, přičemž 2 mikrosatelitové lokusy jsou totožné, a proto jsem nalezla celkem 45 unikátních mikrosatelitových lokusů. Z těchto lokusů pocházelo 43 od druhů z řádu trubkonosí a 2 byly popsány u zástupců řádu dlouhokřídlí. Pro tyto lokusy jsem určila optimální teploty nasedání primerů, časy elektroforetické separace a genotypy 6 nepříbuzných jedinců nesyta indomalajského. Počet alel se pohyboval v rozmezí 25.In my master thesis, I was searching the polymorphic microsatellite loci of the Painted stork (Mycteria leucocephala) and I used the method of cross-species PCR amplification. I tested microsatellite loci, which were polymorfic in species of the order Procellariiformes. In a theoretical part of my thesis, I firstly concisely characterized the order Ciconiiformes, especially the family Ciconiidae. I focused on the genus Mycteria and I described the Painted stork in one chapter. Secondly, I wrote few information about the order Procellariiformes, I provided basic facts about microsatellites and I theoretically described the method of cross-species PCR amplification. Finally, I characterized all microsatellites, which were described in the order Procellariiformes and a couple of microsatellites described in the orders Charadriiformes and Passeriformes. I was looking for the polymorphic microsatellite markers of the Painted stork in the practical part of my master thesis. I used the method of cross-species PCR amplification and I tested overall 212 microsatellite loci, 207 of these loci were described in species of the order Procellariiformes, 4 microsatellite markers were described in species of the order Charadriiformes and 1 microsatellite marker from species of the order Passeriformes. All of these loci were polymorphic in species of the order Procellariiformes. I found overall 46 polymorphic microsatellite markers, two of these markers were identical, so I found 45 unique polymorphic microsatellite loci. From these loci, 43 were described in species of the order Procellariiformes and 2 loci were described in species by the order Charadriiformes. For these microsatellites I identified the optimal annealing temperatures, optimal time of electrophoretic separation and genotypes of 6 unrelated individuals of Painted stork. The number of alelles varied between 2 and 5.
