IgA nefropatie (IgAN) je autoimunitní onemocnění charakterizované ukládáním imunokomplexů (IK) v glomerulárním mezangiu. IK jsou tvořeny polymerními IgA1 molekulami, které mají abnormální strukturu O-vázaných glykanů na pantové oblasti. Pantová oblast IgA1 obsahuje devět potenciálních míst pro O-glykosylaci, ale obvykle jen 3 až 6 míst je obsazených. O-vázané glykany pantové oblasti IgA1 jsou syntetizovány v jasně definovaném sledu enzymatických reakcí. U IgAN dochází k abnormální glykosylaci pantové oblasti IgA1 spočívající ve sníženém množství Gal připojené ke GalNAc. Abnormální glykosylace je příčinou vazby některých protilátek třídy IgG, IgA1, které reagují s abnormálními cukry na pantové oblasti IgA1. Tyto protilátky se uplatňují jako autoprotilátky. Hlavním cílem práce byla produkce rekombinantních enzymaticky aktivních lidských glykosyltransferáz - GalNAc-T2 a -T14, C1GalT1, Cosmc, ST6GalNAc I a II pomocí bakulovirového nebo savčího expresního systému. Rekombinantní glykosyltransferázy umožňují modelovat in vitro průběh glykosylace IgA1 a přispívají tak k poznání molekulární podstaty tvorby abnormálně glykosylovaného IgA1 u IgAN.IgA nephropathy (IgAN) is associated with abnormal O-glycosylation of immunoglobulin A1 (IgA1), characterized by the presence of galactose (Gal)-deficient O-glycans in the IgA1 hinge region (HR). The terminal N-acetylgalactosamine (GalNAc) of IgA1 O-glycans is often modified by addition of sialic acid (NeuAc). IgA1 with terminal GalNAc is recognized by naturally occurring antibodies, thus leading to the formation of pathogenic IgA1-containing circulating immune complexes (CIC) which after glomerular deposition induce mesangial cell proliferation and expansion of extracellular matrix. The aim of this thesis was to study enzymes involved in O-glycosylation of IgA1 hinge region. Recombinant enzymes involved in O-glycosylation of IgA1 hinge region were expressed by the Baculovirus Expression System and/or in HEK 293 cells. Recombinant glycosyltransferases provide valuable tool for elucidation of mechanisms involved in production of aberrantly O-glycosylated IgA1 in IgAN. We hope that this work will contribute to the elucidation of mechanisms involved in generation of aberrantly glycosylated IgA1 in IgA nephropathy.
