Cytoskelet je jednou ze zásadních struktur eukaryotické buňky poskytuje jí oporu, možnost reagovat na vnější podněty a transportovat makromolekulární látky nebo celé organely. Složky rostlinného cytoskeletu mikrotubuly a aktinová mikrofilamenta interagují s množstvím dalších proteinů a také spolu navzájem. Přípravou cytoskeletálních mutantů Arabidopsis thaliana exprimujících vybrané cytoskeletální markery především pro účely studia aktin-mikrotubulárních interakcí se zabývá tato bakalářská práce. Mutantní rostliny exprimující cytoskeletální markery byly získány Agrobacteriem zprostředkovanou transformací rostlin Arabidopsis s mutovaným genem pro AKTIN 2 (mutace je dále označována jako der z anglického deformed root hair) metodou "floral dip" s využitím konstruktů nesoucích cytoskeletální markery pro zviditelnění aktinového a/i mikrotubulárního cytoskeletu. Der1-3 mutantní rostliny Arabidopsis byly také použity jako mateřské rostliny pro křížení s rostlinami exprimujícími cytoskeletální markery. U T1 a T2 generace získaných semen byla stanovena klíčivost a úspěšnost transformace či křížení a vybrané semenáčky byly poté analyzovány s pomocí konfokálního mikroskopu. U studovaných rostlin byl pozorován neporušený aktinový cytoskelet, přestože byla potvrzena přítomnost mutantní alely act2. Mikrotubulární cytoskelet však vykazoval zvýšenou míru neuspořádanosti a depolymerizace. Tyto výsledky naznačují propojení obou složek cytoskeletu.Cytoskeleton is one of the very important part in the eukaryotic cell providing mechanical support, quick reaction to external stimuli and intracellular transport of macromolecules and whole organelles. Microtubules and actin microfilaments, two major components of the pant cytoskeleton, interact both with each other as well as with the large amount of other proteins. Theoretical background of this bachelor thesis is focused on plant cytoskeleton and methods for plant transgenosis. In the practical part, I prepared mutants of Arabidopsis thaliana expressing selected cytoskeletal markers using Agrobacterium mediated transformation and crossing methods. Preliminary microscopical observation were also done on T2 generation of transformats and crossed plants. Mutant plants with expression of cytoskeletal markers were obtained via Agrobacterium-mediated transformation of ACTIN2 (ACT2) mutant Arabidopsis plants (der1-3) using "floral dip" method with constructs carrying cytoskeletal markers to visualize both actin and microtubules. Der1-3 mutant plants were also used as maternal plants for crossing with plants expressing cytoskeletal markers. Transgenic mutant plants of T1 and T2 generation were analyzed in the confocal microscope. Studied plants seemed to have intact actin cytoskeleton despite of confirmed presence of mutant act2 allele. However, microtubular cytoskeleton showed increased bundle formation and disorganization and also inreased depolymerization. These results suggest interconnection between both parts of cytoskeleton.
