Transfekce patří již více než čtyři desetiletí k populárním výzkumným nástrojům pro objasňování genové struktury, regulace a funkce. K vložení cizorodé nukleové kyseliny do buňky lze obecně využít metody biochemické, fyzikální nebo biologické. Z posledně jmenovaných se velké oblibě těší především využití lentivirových vektorů. Tyto vektory se konstruují tak, aby se eliminovala rizika (inzerční mutageneze, onkogeneze) spojená s životním cyklem lentivirů a manipulace s nimi je pak zcela bezpečná. Jednou z řady možností využití lentivirových vektorů je studium buněčné signalizace. V tomto ohledu jsou běžně komerčně dostupné různé systémy umožňující monitoring různorodých signálních drah v buňce. Pro tyto účely jsou lentivirové vektory navrženy a konstruovány tak, že umožňují studium aktivace/inhibice klíčového transkripčního faktoru dané signální dráhy v buňce, kvantitativně měřitelnou prostřednictvím signálu reportéru, nejčastěji světluškové luciferázy. Úspěšné vpravení takto navržených vektorů do buňky umožňuje studovat mechanismus působení různých nízkomolekulárních látek či ligandů. V této práci jsem využila komerčně dostupný lentivirový vektor kódující inducibilní AP-1 (proteinový aktivátor 1) responsivní konstrukt exprimující luciferázu, který byl transdukován do linie potkaního glioblastomu za účelem monitoringu signální dráhy MAPK/JNK (mitogenem aktivovaná protein kináza/c-Jun N-terminální kináza). Tato dráha představuje jeden z downstream cílů RAGE (receptor for advanced glycation end product) signalizace. Na připravené senzorické buněčné linii byl testován účinek vybraných derivátů pyrimidinu jako potenciálních RAGE inhibitorů. RAGE receptor a jeho signalizace jsou zapojeny do patogeneze některých závažných onemocnění. Až na výjimky bývá exprese RAGE při těchto onemocněních zvýšená a proto představuje tento receptor zajímavý terapeutický cíl.Transfection is a popular research tool for clarifying structure, regulation and function of genes. For insertion of foreign nucleic acid into a cell we can use biochemical, physical or biological methods. Very popular is the use of lentiviral vectors. These vectors are designed to eliminate risks associated with the life cycle of lentiviruses (insertional mutagenesis, oncogenesis), so the manipulation is completely safe. One of the many possibilities of using lentiviral vectors is the study of cell signaling. Currently, there are various commercially available systems, which enable monitoring of signal pathways in the cell. For these purposes, lentiviral vectors are designed and constructed to enable the study of activation or inhibiton of a key transcription factor of the signaling pathway in the cell, which is quantitatively measurable through the signal of a reporter (mostly firefly luciferase). Succesful insertion of these vectors into the cell allows us to study the mechanism of action of various small molecules or ligands. In this bachelor´s thesis, I used commercially available lentiviral vector, which encodes inducible AP-1 (protein activator 1) responsive construct expressing the luciferase. This vector was transduce into rat glioblastoma cell line for monitoring of the MAPK/JNK (mitogen-activated protein kinase/c-Jun N-terminal kinase) signaling pathway. This signaling pathway is a downstream target of the receptor RAGE (receptor for advanced glycation end products) signalization. The effect of two pyrimidine derivatives as potential RAGE inhibitors was tested on a prepared sensory cell line. RAGE and its signalization are involved in pathogenesis of some serious diseases. With a few exceptions, the expression of this receptor is increased during these diseases, so it is an interesting therapeutic target.
