Genom eukaryotních buněk je rozdělen mezi jednotlivé chromozomy nacházející se v buněčném jádře. V interfázním jádře vyšších eukaryot však chromozomy nejsou rozmístěny náhodně, ale jsou uspořádány do tzv. chromozomálních teritorií. Studium těchto teritorií se provádí technikou fluorescenční in situ hybridizace metodou tzv. barvení chromozomů a je obvyklá u savců, ptáků, plazů a hmyzu, u rostlin je z důvodu většího výskytu repetitivních sekvencí a větší velikosti genomu tato technika mnohem složitější. V posledních 14 letech se tato technika začala přenášet na genetický model Arabidopsis thaliana z důvodu jeho malého genomu a nízkému počtu repeticí. Zdrojem fluorescenčních sond byla v tomto případě DNA izolovaná z BAC knihoven, nesoucí v jednotlivých liniích Escherichia coli fragmenty studovaného chromozomu. Cílem této bakalářské práce bylo na tuto studii navázat a sestrojit celochromozomovou sondu komplementární k chromozomu 1 Arabidopsis thaliana. To obnáší klonování a izolaci DNA z BAC knihoven, fluorescenční značení próby a její hybridizaci na chromozom 1 v buněčném jádře protoplastů Arabidopsis thaliana a poté sledovat změny v uspořádání chromozomálního teritoria 1 v závislosti odpovědi na stres. Celochromozomová próba byla sestavena a nepřímo fluorescenčně označena. Byla provedena hybridizace na cílovou strukturu v buněčném jádře protoplastů Arabidopsis thaliana, avšak signál nebyl pozorován.The genome of eukaryotic cells is divided among the individual chromosomes located in the nucleus. In the interphase nuclei of higher eukyrotes, chromosomes are not randomly distributed but are in so-called chromosome territories. The study of these territories is carried by technique of fluorescence in situ hybridization by method called "chromosome painting" and is common in studies of mammals, birds, reptiles and insects. This technique is much more complicated in plants due to higher presence of repetitive sequences. In the last 14 years, this technique has begun to permeate to the genetic model plant Arabidopsis thaliana, because of its small genome and low number of repetitive sequences. In this case, the source of probes was DNA isolated from a BAC library, which was carried by a number of clones of Escherichia coli. The objective of this work is to construct whole-chromosome probe complementary to chromosome 1 of Arabidopsis thaliana built on this study. It involves cloning and isolating DNA from BAC libraries, fluorescent labeling of this probe and its hybridization to chromosome 1 in the cell nucleus of Arabidopsis thaliana protoplasts and consequent monitoring of changes in the structure of the chromosome territory 1 depending on the response to a stress. The whole-chromosome probe was constructed and indirectly fluorescently labeled. It was let to hybridize to the target structure in the cell nucleus of Arabidopsis thaliana protoplasts, but the signal was not observed.
