V minulosti se tropickým druhům sinic nevěnovala výrazná pozornost. Preferoval se výzkum převážně temperátní zóny, v posledních letech se zájem vědců soustředil i na polární region. Taxonomické údaje z tropů jsou dosti chudé. V této práci jsou studovány cyanobakteriální kmeny z Bornea, Jávy, Vietnamu, Mongolska a bajkalské části Ruska. Celkově bylo vyizolováno 29 čistých kmenových kultur. Byla provedena morfologická analýza a fylogenetická inference na základě fragmentu genu 16S rRNA. S pomocí polyfázického přístupu jsem byla schopna identifikovat například druh Geitlerinema splendidum, Arthronema africanum, Fischerella cf. tisserantii, která nebyla dosud molekulárně studována. Devět kmenů bylo identifikováno na základě morfologie jako rod Leptolyngbya. Podle analýzy 16S rRNA netvořily koherentní skupiny a nesdílely klastr s Leplyngbya sensu stricto. Je to tedy další důkaz polyfyletického původu tohoto rodu. Byly také determinovány dosud zcela nepopsané rody sinic: candidatus Pinokia longirostra gen. et sp. nov., candidatus Ilonia javanensis gen. et sp. nov., candidatus Motykia elegans gen. et sp. nov. Na základě fylogenetické analýzy byla zjištěna signifikantní podobnost kmenů vyizolovaných z tropů a polárních oblastí. To napovída, že mohou existovat některé druhy sinic s vysokou environmentální tolerancí a velkým geografickým areálem výskytu.The tropical biodiversity of cyanobacteria have been largely overlooked. Researchers preferred the temperate zone or the polar regions. Thus taxonomic data from the tropics are quite scarse. In this work, I studied the cyanobacterial strains from Borneo, Java, Vietnam, Mongolia and the baikal parts of Russia. Altogether 29 clean cultures were isolated. Morphological analysis and phylogenetic inference based on the 16S rRNA gene fragment were performed. Using the polyphasic approach, I was able to identify, for example Geitlerinema splendidum, the Arthronema africanum, and Fischerella cf. tisserantii, which has not yet been molecular studied. Based on a morphology, nine strains were identified as the genus Leptolyngbya. However, according to a analysis of the 16S rRNA, these nine strains did not form a coherent group and they did not share a cluster with the Leptolyngbya sensu stricto. This is therefore further evidence of the polyfyletic origin of this genus. There were also determined completely undescribed genera of cyanobacteria: candidatus Pinokia longirostra gen. et sp. nov., candidatus Ilonia javanensis gen. et sp. nov. and candidatus Motykia elegans gen. et sp. nov. Based on a phylogenetic analysis. I found the significant similarity of strains from tropical and polar regions. It suggests high environmental tolerance and large geographical area of some species.
