Sarkoidóza je zánětlivé, systémové, granulomatózní onemocnění. Ukazuje se, že v patogenezi sarkoidózy hrají důležitou roli genetické faktory. Kandidátní geny mohou být jak predisponující, tak i modifikující toto onemocnění. Ve své diplomové práci jsem se zabývala studiem vybraných jednonukleotidových polymorfismů v genech asociovaných se sarkoidózou a jejich využitelností k predikci vývoje onemocnění. Ke studiu byla využita DNA izolovaná z periferní krve pacientů. Soubor tvořilo 123 pacientů s plicní sarkoidózou, u kterých byl znám vývoj onemocnění po 2 letech. Genotypizace 5 polymorfismů rs2076530 (BTNL2), rs1049550 (ANXA11), rs916977 (HERC2), rs11209026 (IL-23R) a rs1040461 (RAB23) byla provedena pomocí technologie MassArray. Mezi skupinami pacientů, u kterých došlo k regresi a progresi onemocnění, nebyly nalezeny signifikantní rozdíly v genotypových a alelických frekvencích. Nepodařilo se objevit žádné genetické markery, které by mohly být využity k predikci vývoje onemocnění a identifikaci pacientů, u kterých dochází k progresi onemocnění.Sarcoidosis is an inflammatory, systemic granulomatous disease. The genes play a pivotal role in the pathogenesis of sarcoidosis. These genes can be predisposing as well as modifying the disease. The aim of this thesis was to assess suitability of the candidate single nucleotide polymorphisms in genes associated with sarcoidosis for the prediction of disease development. DNA was obtained from peripheral blood of 123 patients with sarcoidosis. The patients were divided according to the disease development after 2-years. Five single-nucleotide polymorphisms associated with sarcoidosis and its clinical phenotypes, namely rs2076530 (BTNL2), rs1049550 (ANXA11), rs916977 (HERC2), rs11209026 (IL-23R), rs1040461 (RAB23) were tested using MassArray technology (Sequenom). We did not detect any significant difference in allele frequencies for all studied polymorphisms. Also the distribution of genotype frequencies did not differ between patients with regression and progression of disease. In conclusion, none of studied polymorphisms may serve as a genetic marker suitable for the prediction of the disease development, and thus identify patients with the progressive disease status.
