Olomoucin II a látka NGF1568 patří mezi perspektivní syntetické látky odvozené od rostlinných cytokininů, které by mohly v budoucnosti sloužit k terapii nádorových onemocnění či zánětlivých stavů. Doposud neexistují informace o interakcích těchto potenciálních léčiv s významnými lidskými enzymovými systémy podílejícími se na metabolismu xenobiotik. Předkládaná disertační práce je v první části zaměřena na studium inhibičního vlivu olomoucinu II a NGF1568 na enzymové aktivity hlavních enzymů I. fáze metabolismu xenobiotik, cytochromů P450 (CYP). V další části disertační práce byl studován potenciální metabolismus olomoucinu II a NGF1568 biotransfromačními enzymy obsaženými v jaterní mikrosomální frakci [(CYP) a UDP-glukuronosyltransferasy] a cytosolickými sulfotransferasami.Olomoucine II and substance NGF1568 belong to prospective synthetic compounds derived from plant cytokinines with potential use in cancer therapy (olomoucine II) or as antiinflammatory agents (NGF1568) in the future. The interactions of these potential drugs with human enzyme systems participating in drug metabolism have not been evaluated yet. The first part of thesis presented here is focused on study of inhibitory effect of olomoucine II and NGF1568 on enzymatic activities of main phase I drug metabolising enzymes, cytochromes P450 (CYP). In the next part, the potential metabolism of olomoucine II and NGF1568 by human liver microsomal enzymes of drug metabolism (CYP and UDP-glucuronosyltransferases) and by human liver cytosolic sulfotransferases was studied.
