DNA-proteinové crosslinky jsou jedno z nejškodlivějších DNA poškození a pokud nedojde k jejich včasné opravě, může dojít k narušení buněčných procesů a následnému ohrožení životaschopnosti buněk. Proto si organismy vytvořily hned několik specializovaných opravných drah. Právě proteiny WSS1A a STUbL2 jsou spojeny s opravami DNA-proteinových crosslinků, ale jejich mechanismus u rostlin zatím nebyl dostatečně prozkoumán. Pro lepší pochopení opravných mechanismů je třeba najít nové proteiny, které se na nich spolupodílejí. Pro sledování proteinových interakcí byla nedávno vyvinuta nová metoda tzv. proximity labeling, která umožňuje pomocí vybraných enzymů modifikovat proteiny interagující s vybraným studovaným proteinem nebo ležící v jeho blízkosti. Jedním takovým enzymem je i TurboID. Označené proteiny se následně dají purifikovat a analyzovat např. pomocí hmotnostní spektrometrie. Takto je možné např. identifikovat nové interaktory kandidátních proteinů. Díky tomu lze získat nové informace sloužící k bližšímu pochopení opravných mechanismů DNA-proteinových crosslinků u rostlin. Pomocí Gateway technologie klonování byly vytvořeny konstrukty nesoucí kódující sekvenci genu WSS1A a TurboID tag. Tyto konstrukty byly transformovány do rostlin Arabidopsis thaliana a byly vyselektovány transgenní rostliny obsahující vložený konstrukt. Tyto budou dále sloužit pro následnou analýzu interaktorů WSS1A pomocí TurboID afinitní purifikace.DNA-protein crosslinks are one of the most harmful types of DNA damage, and if they are not repaired in time, cellular processes may be disrupted and cell viability may be threatened. Therefore, organisms have created several specialized repair pathways. It is the WSS1A and STUbL2 proteins that are associated with the repair of DNA-protein crosslinks, but their mechanism has not been sufficiently explored in plants yet. To better understand the repair mechanisms, we need to find new proteins, that are involved. To identify protein-protein interactions, a new method called proximity labeling has recently been developed, which allows using enzymes to identify proteins interacting with the protein of interest or proteins in the vicinity of it. For example, TurboID is one of the enzymes that are used for proximity labeling method. Such labeled proteins can be purified and further analysed. Therefore, new information can be obtained, for example for betterunderstanding of the repair mechanisms of DNA-protein crosslinks in plants. Using Gateway cloning technology, constructs carrying the coding sequence of the WSS1A gene and the TurboID tag were generated. These constructs were transformed into Arabidopsis thaliana plants and transgenic plants containing the inserted construct were selected. These will be further used for subsequent analysis of WSS1A proximal interactome by TurboID affinity purification.
