Tato diplomová práce se zabývá enzymem polyfenoloxidasou, jeho účastí v obranném metabolismu rostlin a jeho expresí in vitro. V teoretické části jsem popsala genomickou organizaci genů pro polyfenoloxidasu v rostlinách, strukturu tohoto enzymu, jeho katalytickou funkci a jeho uplatnění v potravinářském a kosmetickém průmyslu. Cílem praktické části bylo exprimovat tento protein z planého hrachu kultivaru JI64 v buňkách Escherichia coli a tento protein biochemicky charakterizovat. Gen pro polyfenoloxidasu byl amplifikován pomocí PCR a vložen do plasmidu obsahujícího GST kotvu. Tento konstrukt byl po ověření agarosovou elektroforézou a osekvenování transformován do buněk E. coli pro expresi BL21 StarTM (DE3). Po indukci exprese přidáním IPTG byl enzym purifikován pomocí GST afinitní chromatografie. Jeho přítomnost a čistota byla ověřena pomocí SDS-PAGE. Spektrofotometricky byla stanovena jeho specifická aktivita, pH optimum, počet atomů mědí ve struktuře, optimální koncentrace SDS pro aktivaci latentní formy a vliv inhibitorů na jeho aktivitu. Studovaly se reakce s 12 vybranými potencionálními substráty. Získaný enzym se specifickou aktivitou 29,545 U*mg-1 byl zařazen mezi tyrosinasy. Obsahoval 3 atomy mědi a jeho pH optimum bylo 6,00. Vhodná koncentrace SDS pro aktivaci enzymu byla 0,25 mmol*l-1. Nejlepším substrátem byl adrenalin. Nejúčinnějšími inhibitory enzymu byl tropolon a thujaplicin. Jedná se o první známou expresi polyfenoloxidasy ze semen hrachu a popis jejích biochemických vlastností.This diploma thesis deals with study of polyphenol oxidase enzyme, its role in defense metabolism of plants and with its in vitro expression. The theoretical part deals with the structure of this enzyme, its catalytic function and also genomic organization of its genes. The goal of practical part was to express this protein from wild pea cultivar JI64 in the Escherichia coli bacterial expression system and characterize this protein biochemically. The gene for this enzyme was amplified by PCR and inserted into a plasmid as GST tagged construct. After verification by agarose electrophoresis and sequencing, this construct was transformed into E. coli BL21 StarTM (DE3) expression cells. After expression induction by IPTG, the enzyme was purified via GST affinity chromatography. Its presence and purity were verified by SDS-PAGE. Its specific activity, pH optimum, the number of copper atoms in its structure, the optimal concentration of SDS for the activation of its latent form and the effect of inhibitors on its activity were determined spectrophotometrically. Its reactions with 12 different ones were also observed. The obtained enzyme with a specific activity of 29,545 U*mg-1 was included among tyrosinases. This polyphenol oxidase contained 3 copper atoms and its pH optimum was 6,00. The optimal SDS concentration for enzyme activation was 0,25 mmol*l-1. Adrenaline was the best substrate. The most effective inhibitors of the enzyme were tropolone and thujaplicin. This is the first report of recombinant expression of polyphenol oxidase from pea and determination of its biochemical properties.
