Lipoxygenasy jsou enzymatická rodina, která se vyskytuje v živočišné i rostlinné říši. Tyto enzymy katalyzují dioxygenaci polynenasycených mastných kyselin. V jarním ječmeni najdeme dvě lipoxygenasy - LOX-1 a LOX-2. Oba enzymy se liší ve svém účinku v tvorbě derivátů polynenasycených mastných kyselin. Dioxygenovány jsou primárně kyselina linolová a linolenová. Z jejich hydroperoxidů pak vznikají látky trans-2-nonenal (T2N) a trihydroxyoktadekenová kyselina (THOD), jenž mají negativní vliv na chuť a stabilitu pěny piva. Šlechtění odrůd se sníženou či nulovou lipoxygenasovou aktivitou je tedy velmi atraktivní z pohledu pivovarnického průmyslu. Cílem této práce bylo charakterizovat transgenní rostliny generace T1 jarního ječmene (Hordeum vulgare) odrůdy Golden Promise, u nichž je předpokládána mutace v genu LOX-1, která byla vyvolána pomocí technologie CRISPR/Cas9. Byl proveden PCR screening T1 generace a identifikovány transgenní rostliny nesoucí kazetu s sgRNA. Cílová sekvence v genu LOX-1 byla amplifikována a charakterizována Sangerovým sekvenováním. Analýzou získaných sekvencí bylo zjištěno, že transgenní rostliny v T1 generaci nenesly mutaci v genu LOX-1.Lipoxygenases are an enzymatic family that occurs in both the animal and plant kingdoms. These enzymes catalyze the dioxygenation of polyunsaturated fatty acids. In barley we can find two lipoxygenases - LOX-1 and LOX-2. Both enzymes differ in their effect in the formation of polyunsaturated fatty acid derivatives. Linoleic acid and linolenic acid are primarily dioxygenated. Their hydroperoxides then produce trans-2-nonenal (T2N) and trihydroxyoctadecenoic acid (THOD), which have a negative effect on the taste and stability of beer foam. Breeding varieties with reduced or no lipoxygenase activity is therefore very attractive from the perspective of the brewing industry. The aim of this study was to characterize transgenic plants of the T1 generation of barley (Hordeum vulgare) Golden Promise variety, which are expected to be mutated in the LOX-1 gene, which was induced by CRISPR/Cas9 technology. PCR screening of T1 generation was performed and transgenic plants carrying the sgRNA cassette were identified. The target sequence in the LOX-1 gene was amplified and characterized by Sanger sequencing. Analysis of the obtained sequences showed that transgenic plants in the T1 generation did not carry a mutation in the LOX-1 gene.
