Androgenní receptor (AR) patří mezi jaderné receptory a hraje klíčovou roli ve vývoji rakoviny prostaty. Působí jako transkripční faktor pro expresi proteinů, jakými jsou například PSA a Nkx3.1. V případě rakoviny prostaty se nejmodernější vědecké přístupy zaměřují na inhibici transkripční aktivity AR, zablokování vazby ligandu či inhibici koaktivátorů AR, které jsou klíčové pro jeho konečnou aktivitu. Zajímavých úspěchů se v současnosti povedlo dosáhnout ve vývoji technologie PROTAC, která umožňuje cílenou degradaci AR. V praktické části této práce je pozornost zaměřena na studium dvou sérií látek odvozených od galeteronu a jejich vlivu na AR a estrogenní receptor (ER). U vybraných prostatických nádorových linií byl stanoven vliv studovaných látek na transkripční aktivitu AR a pomocí imunoblotingu byly sledovány exprese klíčových proteinů signalizace AR. Součástí práce byla i optimalizace reporterové eseje pro ER pomocí přechodné transfekce s cílem otestovat specifitu vybraných sloučeniny vůči ER.The androgen receptor (AR) is a nuclear receptor and plays a key role in the development of prostate cancer. It acts as a transcription factor for the expression of proteins such as PSA and Nkx3.1. Recent scientific approaches to prostate cancer treatment have focused on inhibiting the AR transcriptional activity, blocking ligand binding or inhibiting AR coactivators which are critical for its ultimate activity. An interesting approach has been introduced by PROTAC technology, which allows for targeted degradation of AR. In the practical part of this thesis, the focus is on the study of two series of compounds derived from galeterone and their influence on AR and estrogen receptor (ER). The effect of the studied compounds on the transcriptional activity of AR was determined in selected prostate cancer cell lines and the expression of key proteins of AR signaling was analysed using immunoblotting. The thesis also includes the optimization of a reporter assay for ER through transient transfection to test the specificity of selected compounds against ER.
