Mutantní typy rostlin s nedostatkem chlorofylu b jsou často využívány ve vědeckých experimentech k výzkumu fotosyntetického aparátu. Chlorofyl b má vliv na množství procesů probíhajících v rostlině, mimo jiné ovlivňuje efektivitu a stavbu fotosyntetického aparátu. V diplomové práci byly na základě experimentálních dat srovnávány rostliny ječmene setého (Hordeum vulgare L., cv. Bonus) a jeho mutantní formy chlorina f2f2, která je specifická nedostatkem chlorofylu b. Hodnoceny byly obsahy pigmentů, fluorescenční parametry chlorofylu a, redoxní stav P700 a plastocyaninu a elektroforetické separace proteinových komplexů tylakoidních membrán. Analýza pigmentů ukázala, že chlorina má snížené množství chlorofylů a a b a karotenoidů oproti přirozenému genotypu ječmene. Na základě hodnocení fluorescenčních parametrů velmi rychlé fluorescenční indukce bylo zjištěno, že chlorina má nižší účinnost fotochemie fotosystému II a menší zastoupení světlosběrných antén než přirozený genotyp ječmene setého. Tyto výsledky odpovídaly zjištěné limitaci účinnosti fotochemie fotosystému I u chloriny na donorové straně elektronového transportního řetězce. Z výsledků měření zhášecí a relaxační analýzy vyplynulo, že chlorina má jinou kinetiku zhášecích procesů než přirozený genotyp a že zhášecí procesy se na zpracování energie podílejí odlišným způsobem. Z elektroforetických separací pigment-proteinových komplexů tylakoidních membrán bylo zřejmé, že chlorina netvoří superkomplexy fotosystému II a světlosběrných komplexů II, které byly typické pro přirozeně se vyskytující genotyp ječmene. U chloriny bylo zaznamenáno menší množství světlosběrných komplexů a odlišné složení pigment-proteinových komplexů.Chlorophyll b deficient mutants are often used in scientific experiments to investigate the photosynthetic apparatus. Chlorophyll b affects a number of processes taking place in plants, including the efficiency and structure of photosynthetic apparatus. Based on experimental data, plants of barley (Hordeum vulgare L., cv. Bonus) and its mutant chlorina f2f2 were compared in the diploma thesis. Pigment contents, parameters of chlorophyll fluorescence, and redox state of P700 and plastocyanin, and electrophoretic separation of protein complexes of the thylakoid membranes were evaluated. Analysis of pigments showed that chlorina had a reduced content of chlorophyll a and b and carotenoids compared to wild type barley. Based on the fluorescence parameters of very fast fluorescence induction, it was found that chlorina has lower efficiency of the photochemistry of photosystem II and smaller amount of the light-harvesting complexes than the natural genotype of barley. These results corresponded to the observed limitation in electron transfer in photosystem I donor side in chlorina. The results of quenching and relaxation analysis showed that chlorina had different quenching kinetics than wild type and that the quenching processes participated differently in energy dissipation. From the electrophoretic separations of pigment-protein complexes of thylakoid membranes, it was clear that chlorina did not form supercomplexes made of photosystem II and light-harvesting complex II, which are typical in the naturally occurring barley genotype. For chlorina, smaller amounts of light-harvesting complexes and different compositions of pigment-protein complexes were noted.
