Bakalářská práce se zabývá viry infikujícími brukvovitou zeleninu (r. Brassica) se zaměřením na Turnip mosaic virus (TuMV) a jeho hostitelský okruh. V teoretické části práce jsou popisovány hlavní viry rostlin rodu Brassica se zaměřením na jejich taxonomické zařazení, geografické rozšíření, hostitelský okruh, příznaky infekce, přenos a šíření a molekulární vlastnosti. Konkrétně se jedná o Cauliflower mosaic virus, Turnip yellow mosaic virus a Turnip yellows virus. Zvláštní důraz je kladen na Turnip mosaic virus, u kterého jsou jednotlivé vlastnosti popsány podrobněji, charakterizována je také genetická variabilita a metody detekce. Závěr teoretické části je věnován metodě kvantitativní PCR a jejímu využití v detekci rostlinných virů. Experimentální část byla zaměřena na detekci a následnou charakterizaci získaných izolátů TuMV. Pomocí sekvenování a fylogenetické analýzy byla zjištěna genetická variabilita 11 izolátů TuMV pocházejících z různých oblastí České republiky. Dále byla vyvinuta a optimalizována metoda detekce TuMV pomocí RT-kvantitativní PCR, která zahrnovala navržení primerů, optimalizaci teploty nasedání primerů a koncentrace primerů a absolutní kvantifikaci s využitím SYBR Premix Ex TaqTM.This bachelor thesis deals with viruses of Brassicaceae vegetable plants (g. Brassica) with the focus on the Turnip mosaic virus and its host range. In the theoretical part the main viruses of the genus Brassica are described with the focus on their taxonomic classification, geographical distribution, host range, infection symptoms, transmission and spread and molecular properties. Specifically, it is Cauliflower mosaic virus, Turnip yellow mosaic virus and Turnip yellows virus. Special emphasis is placed on the Turnip mosaic virus, in which the properties are described in more detail and the genetic variability and detection methods are also characterized. The end of the theoretical part is devoted to the quantitative PCR method and its use in the detection of plant viruses. The experimental part was focused on detection and subsequent characterization of obtained TuMV isolates. Sequencing and phylogenetic analysis revealed the genetic variability of 11 TuMV isolates from different regions of the Czech Republic. The TuMV detection method was developed and optimized using RT-quantitative PCR, which included primer design, optimization of primer annealing temperature and primer concentration and virus absolute quantification using SYBR Premix Ex TaqTM.
