Předložená diplomová práce se věnuje studiu interakcí proteinů s ligandy pomocí hmotnostní spektrometrie. V teoretické části je zpracována literární rešerše na téma hmotnostní spektrometrie a protein-ligandových interakcí. Experimentální část práce se zabývá stanovením optimálního koncentračního rozsahu cytochromu c a hovězího sérového albuminu a následně měřením interakcí obou proteinů s ligandy. Pro měření byly použity dva hmotnostní spektrometry ESI/QqTOF a MALDI-TOF. Jako ligandy byly zvoleny Triterpen, karboran, tryptofan a cisplatina. Vyvinutá metoda byla použita ke studiu interakce avidinu s modifikovaným biotinem.The thesis is devoted to the study of interactions between proteins and ligands using mass spectrometry. The theoretical part includes the literature review of mass spectrometry and protein-ligand interactions. The experimental part deals with the determination of the optimum concentration range of cytochrome c and bovine serum albumin and analysis of the interaction of above mentioned proteins with ligands. Two mass spectrometers ESI/QqTOF and MALDI-TOF were used in this analysis. Triterpene, carborane, tryptophan and cisplatin were chosen as ligands. Developed method was used for verification of interaction of avidin and modified biotin.
