Cytochromy P450 hrají důležitou roli v metabolismu xenobiotik a hormonů. Ze strukturního pohledu jde o enzymy se zanořeným aktivním místem, ke kterému substráty přistupují pomocí tunelů. Tyto tunely mohou hrát roli pro selektivitu jednotlivých cytochromů P450, které jsou na jednu stranu vysoce specializované v místě metabolismu jednotlivých látek, ale současně značně promiskuitní, co se výběru substrátů týče. V této práci se zaměřuji na studium mechanismu vstupu cholesterolu do aktivního místa cytochromu P450 11A1 s použitím molekulárních simulací. Cílem této práce je seznámit čtenáře s metabolismem cholesterolu a ukázat na možné mechanismy vstupu cholesterolu do prvního enzymu na metabolické dráze mitochondriálního cytochromu P45011A1. Nakonec jsem vstup cholesterolu jakožto modelového substrátu popsala simulačními technikami a poukázala na další možné použití těchto technik.Cytochromes P450 play an important role in the metabolism of xenobiotics and hormones. From a structural point, they are enzymes with an internalized active site, which is accessible for substrates through tunnels. These tunnels may play a role in the selectivity of the individual cytochromes P450, as they are extremely specific about the placement of the substrate metabolism, while at the same time being quite promiscuous with regards to substrate choice. This thesis focuses on the study of the mechanism of entry of cholesterol into the cytochrome P450 11A1 active site, using molecular simulations. The aim of this thesis is to introduce the reader to the metabolism of cholesterol problematic and to illustrate the possible mechanisms of entry of cholesterol into the first enzyme of the metabolic pathway mitochondrial cytochrome P450 11A1. In addition, the entry of cholesterol, as a model substrate, was described using simulation techniques, and other potential applications of these techniques were illustrated.
