Tato bakalářská práce se zabývá hledáním polymorfních mikrosatelitů u plameňáka karibského (Phoenicopterus ruber) za pomocí cross-species PCR amplifikace mikrosatelitních lokusů izolovaných od zástupců řádu trubkonosí. Teoretická část je zaměřená na problematiku zařazení plameňáka karibského do systému a změny, kterými toto zařazení v průběhu času procházelo, popisuje řád plameňáci, čeleď plameňákovití a také plameňáka karibského. Dále se věnuje obecné charakteristice mikrosatelitů, výskytu mikrosatelitů v ptačím genomu a popisu de novo a cross-species mikrosatelitů pro plameňáka karibského a mikrosatelitů izolovaných od druhů z řádu trubkonosí. V experimentální části byla na genomické DNA šesti nepříbuzných jedinců plameňáka karibského otestována cross-species PCR amplifikace celkem 213 mikrosatelitů, z čehož 207 představovalo všechny de novo mikrosatelity doposud odvozené od zástupců z řádu trubkonosí a u zbylých 6 se jednalo o cross-species mikrosatelity polymorfní u zástupců z řádu trubkonosí, původně izolované u zástupců z řádu dlouhokřídlí a pěvci. Všechny testované mikrosatelity poskytly monomorfní nebo polymorfní produkt a celkem bylo nalezeno 50 polymorfních mikrosatelitů.This bachelor thesis deals with the searching for polymorphic microsatellites in the Caribbean flamingo (Phoenicopterus ruber) using the method of cross-species PCR amplification of microsatellite loci isolated from representatives of the order Procelariiformes. The theoretical part of the thesis is focused on the problematic of the taxonomic classification of the Caribbean flamingo and its changes in the course of time, and it also describes the order Phoenicopteriformes, family Phoenicopteridae and the Caribbean flamingo. Furthermore, it characterizes microsatellites and their occurrence in the avian genome, and finally, it discusses de novo and cross-species isolated microsatellites in the Caribbean flamingo and microsatellites derived from species of the order Procelariiformes. In the experimental was performed the cross-species PCR amplification of 213 microsatellites, which included all 207 so far known loci isolated from representatives from the order Procellariiformes and 6 cross-species microsatellites polymorphic in the order Procellariiformes isolated from representatives from the order Charadriiformes and Passeriformes, on the genomic DNA of six unrelated individuals of the Caribbean flamingo. All of the tested microsatellites provided monomorphic or polymorphic products and in total 50 polymorphic microsatellites were found.
