V rámci teoretické části byl vypracován přehled zaměřený na vlastnosti transportních proteinů ABCG5/8 a možnosti jejich ovlivnění. Práce se dále zabývá metabolickým syndromem a potkany s dědičnou hypertriglyceridémií, které jsou akceptovaným modelem metabolického syndromu. Jako podpůrná léčba metabolického syndromu by se mohl použít silymarin, tedy přírodní extrakt z ostropestřce mariánského, nebo omega-3 polynenasycené mastné kyseliny. Víme, že tyto přírodní látky zvyšují expresi proteinů ABCG5/8 v játrech hypertriglyceridemických potkanů. Cílem této práce bylo potvrdit, zda silymarin, omega-3 polynenasycené mastné kyseliny a jejich kombinace ovlivňují expresi i střevních transportérů ABCG5/8 na úrovni proteinu u potkaního modelu metabolického syndromu.Z výsledků vyplývá, že potkani na dietě obohacené o vybrané přírodní látky neměli změněnou expresi střevních ABCG5/8 v porovnání s kontrolními potkany, kteří byli krmeni standardní dietou bez obsahu silymarinu a omega-3 polynenasycených mastných kyselin. Výsledky tedy nepotvrdily efekt v játrech potkanů. Důvodů může být několik, např. mohou zde hrát velkou roli střevní bakterie nebo naopak v játrech metabolity vzniklé ze silymarinu a omega-3 polynenasycených mastných kyselin. K vysvětlení rozdílných účinků vybraných přírodních látek na expresi ABCG5/8 v játrech a ve střevech je zapotřebí provést další experimenty, které jsou v budoucnu v plánu.The summary focused on qualities of transport proteins ABCG5/8 and possibilites of their influencing has been elaborated in the theoretical part. Then the study deals with a metabolic syndrome and rats with hereditary hypertriglyceridemia which are an accepted model of the metabolic syndrome. Silymarin (natural extract of milk thistle) could be used as a supporting therapy of the metabolic syndrome or omega-3 polyunsaturated fatty acids. It is known that these natural substances increase the expression of proteins ABCG5/8 in the hypertriglyceridemic rats livers. The aim of this work was to confirm if silymarin, omega-3 polyunsaturated fatty acids and their combinations influence the expression also at intestinal transporters ABCG5/8 on a protein level at rats model of the metabolic syndrome. From the results it arises that the rats which were on a diet enriched by chosen natural substances did not have different expression intestinal ABCG5/8 in comparison with the inspecting rats which were on a standard diet without the contents of silymarin and omega-3 polyunsaturated fatty acids. The results did not confirm any effect in the rats intestines. There can be several reasons for this fact, e.g. some intestinal bacterias can play a big role or on the contrary metabolites which have arisen from silymarin in the livers and omega-3 polyunsaturated fatty acids. Some other experiments (which are planned in the future) are needed to be done for the explanation of different effects of chosen natural substances for the expression ABCG5/8 in livers and intestines.
