Uvedená diplomová práce je zaměřena na studium patofyziologie Diamondovy-Blackfanovy anémie (DBA). V teoretické části je stručně popsána biogeneze ribozomů, proces erytropoézy a jeho regulace, proces syntézy hemoglobinu a patologie červené krevní řady se zaměřením na anémie a jejich klasifikaci. Důraz je kladen především na DBA, a to konkrétně na její etiologii a dědičnost, klinické projevy, diagnostiku, léčbu a patofyziologii. V experimentální části práce byl použitý buněčný model s vnesenými mutacemi v genech kódujících ribozomální protein l5 (Rpl5) a s19 (Rps19), které jsou nejčastěji mutovanými geny u DBA. Cílem práce bylo ověření vhodnosti vytvořených Rpl5- a Rps19-mutantních klonů pro studium patofyziologie DBA. Analyzována byla exprese Rpl5 a Rps19 genů na úrovni mRNA a proteinů, proliferační kapacita Rp-mutantních buněk, aktivace p53 a exprese erytroidního transkripčního faktoru Gata1. Výsledky uvedených metod potvrdily sníženou produkci Rpl5 a Rps19, sníženou expresi Gata1 mRNA, sníženou proliferaci a zvýšenou aktivaci p53 u Rp-mutantních buněk ve srovnání s buňkami kontrolními. Dále byla provedena analýza hladin reaktivních forem kyslíku (ROS), která ukázala jejich zvýšenou hladinu u Rp-mutantních buněk a z toho plynoucí oxidační poškození DNA. Práce ukázala vhodnost vytvořených buněčných modelů pro studium DBA a také zapojení oxidačního stresu do patofyziologie tohoto onemocnění.The diploma thesis is focused on the study of the pathophysiology of Diamond-Blackfan anemia (DBA). The theoretical part briefly describes the biogenesis of ribosomes, the process of erythropoiesis and its regulation, synthesis of hemoglobin and the pathology of red blood cells with a focus on anemia and their classification. Emphasis is placed primarily on DBA, specifically on its etiology and inheritance, clinical symptoms, diagnosis, treatment and pathophysiology. In the experimental part of the work, cellular models with engineered mutations in the two most commonly mutated genes in DBA, encoding ribosomal protein l5 (Rp15) and s19 (Rps19), were used. The aim of the work was to verify whether the generated Rp15- and Rps19-mutant clones recapitulate the DBA phenotype. The expression of Rp15 and Rps19 genes at the mRNA and protein level was analyzed first, followed by the cell proliferation assay, assessment of p53 activation, and expression analysis of the erythroid transcription factor Gata1. These assays confirmed reduced production of Rp15 and Rps19, reduced expression of Gata1 mRNA, reduced proliferation capacity and increased activation of p53 in Rp-mutant cells compared to control cells. Subsequently, an analysis of the levels of reactive oxygen species (ROS) was performed and showed their increased levels in Rp-mutant cells than in control cells. Furthermore a consequent oxidative DNA damage was detected in Rp-mutant cells. Altogether the work showed the suitability of the created cellular models for studying DBA and also the involvement of oxidative stress in the pathophysiology of this disease.
