Tato práce se zabývá studiem aktivního místa aldehyddehydrogenasy 21 (ALDH21) z mechu Physcomitrella patens. Teoretická část je zaměřena na rešerši současných poznatků o mechu P. patens a nadrodině rostlinných aldehyddehydrogenas. Praktická část zahrnuje místně-řízenou mutagenezi genu PpALDH21, heterologní expresi mutantních forem v Escherichia coli a purifikaci rekombinantních proteinů afinitní chromatografií. Čistota proteinů byla ověřena pomocí SDS-PAGE a sekundární struktura byla ověřena pomocí CD-spektroskopie. U jednotlivých variant PpALDH21 byly následně spektrofotometricky testovány změny v substrátové specificitě, kinetických parametrech a teplotní stabilitě. Výsledky ukazují na význam jednotlivých aminokyselinových reziduí v aktivním místě pro správnou katalytickou funkci enzymu a oxidaci preferovaného substrátu.The thesis is focused on a study of the active site of aldehyde dehydrogenase 21 (ALDH21) from the moss Physcomitrella patens. The theoretical part summarizes the present knowledge of moss P. patens and plant aldehyde dehyderogenase superfamily. The practical part deals with site-directed mutagenesis of PpALDH21 gene, heterologous expression of the mutant variants in Escherichia coli and purification of recombinant proteins by affinity chromgatography. Purity of the final proteins was verified by SDS-PAGE and proper protein folding was confirmed by CD spectroscopy. Each PpALDH21 variant was subsequently spectrophotometrically assayed for changes in its substrate specificity, kinetic parameters and thermal stability. The results show the importance of individual amino acid residues in the active site for a proper catalytic function of the enzyme and oxidation of the preferred substrate.
