Předkládaná diplomová práce je zaměřena na studium dvou mutantních forem S nitrosoglutathionreduktasy (GSNOR) z rostlinného patogena Phytophthora infestans. GSNOR je klíčový regulátor metabolismu S nitrosothiolů. V buňce plní funkci v obranné reakci na biotické i abiotické stresové faktory, podílí se na kontrole homeostázy a chrání organismus před nitrosačním stresem. Teoretická část práce je věnována nejnovějším poznatkům o nitraci a S nitrosylaci proteinů se zaměřením na úlohu a vlastnosti S nitrosoglutathionreduktasy, místně řízené mutagenezi a metodám studia rekombinantních/mutantních proteinů. V experimentální části byla provedena místně řízená mutageneze genu PiGSNOR, a to jednobodové mutace T95Q, S324T a I299V, a jejich následná heterologní exprese v E. coli. Rekombinantní protein PiGSNOR WT a jeho mutantní formy byly purifikovány afinitní chromatografií. Správnost složení proteinů byla ověřena pomocí CD spektroskopie. Další část byla zaměřena na biochemickou charakterizaci proteinů. Byly stanoveny základní kinetické parametry jako je Km, Vlim, skladovací stabilita, pH optimum a IC50 vybraných inhibitorů. Získané výsledky byly porovnány se studií mutantů GSNOR z rostliny Solanum lycopersicum jakožto hostitele P. infestans pro důkladnější porozumění mechanismů interakce rostlina-patogen a k lepšímu pochopení role S nitrosylace a S nitrosoglutathionreduktasy.The diploma thesis is focused on the study of two S-nitrosoglutathione reductase mutant forms from the plant pathogen Phytopthora infestans. The S nitrosoglutathione reductase (GSNOR) is a key regulator of metabolism of S nitrosothiols. In the cell, it performs the function in a defense response to biotic and abiotic stress factors. It participates in the control of the homeostasis and protects the organism from nitrosative stress. The theoretical part of the work is dedicated to the latest knowledge about the nitration and nitrosylation of proteins focusing on the role and properties of the GSNOR, site-directed mutagenesis and methods of study recombinant/mutant proteins. In the experimental part there was realized site directed mutagenesis of PiGSNOR gene, namely single-point mutations T95Q, S324T, I299V, and subsequent heterologous expression in E. coli. Recombinant proteins PiGSNOR-WT and its mutants were purrified by affinity chromatography. The results were verified by CD spectroscopy. Another part was focused on the biochemical characterization of the proteins. Basic kinetic parameters have been determined as Km, Vlim, storage stability, pH optimum, and IC50. The obtained results were compared with the study of GSNOR mutants from Solanum lycopersicum.
