V této bakalářské práci jsem se zabývala hledáním polymorfních mikrosatelitů u nesyta afrického (Mycteria ibis) metodou cross-species PCR amplifikace. V teoretické části jsem se věnovala taxonomickému zařazení nesyta afrického. Popsala jsem řád brodiví (Ciconiiformes), čeleď čápovití (Ciconiidae) a rod nesyt (Mycteria). Následovala také kapitola týkající se nesyta afrického. V další části rešerše jsem se věnovala popisu repetitivní DNA. Nejvíce jsem se zabývala mikrosatelity, jejich rozdělením, využitím a hledáním. Poslední část teoretického úvodu jsem věnovala popisu hledání mikrosatelitových lokusů navržených pro druhy z řádů brodiví, potáplice a potápky. V praktické části jsem se zaobírala testováním mikrosatelitových lokusů metodou cross-species PCR amplifikace u 6 jedinců nesyta afrického, jehož vzorky krve byly získány ze ZOO Zlín-Lešná. Celkem bylo testováno 219 párů primerů. Z řádů brodiví a potáplice byly testovány všechny dosud nalezené mikrosatelitové lokusy, z řádů potápky a vrubozobí pouze některé vybrané mikrosatelity. Polymorfní produkt jsem zaznamenala u 63 párů primerů, z nichž 60 bylo původně navrženo pro ptáky z řádu brodiví. 1 polymorfní mikrosatelit pocházel od potáplice lední (řád potáplice) a 2 od kachny divoké (řád vrubozobí). Počet alel se pohyboval od 2 do 8.In this bachelor thesis I engaged in search of polymorphic microsatellites in yellow-billed stork (Mycteria ibis) by cross-species PCR amplification. In the theoretical part I dealt with taxonomic classification of the yellow-billed stork. I described the order Ciconiiformes, family Ciconiidae and genus Mycteria. Followed by a chapter about yellow-billed stork. In the next part of the research I focused on the description of repetitive DNA. Mainly I dealt with microsatellites, their distribution, use and search. In the last part of the theoretical introduction I focused on description of searching microsatellite loci designed for species of orders Ciconiiformes, Gaviiformes and Podicipediformes. In the experimental part I engaged in testing their microsatellite loci by cross-species PCR amplification on 6 individuals of yellow-billed stork. These blood samples were obtained from ZOO Zlín-Lešná. Altogether, 219 pairs of primers were tested. For orders Ciconiiformes and Gaviiformes all found microsatellite loci were tested, for the orders Podicipediformes and Anseriformes only some microsatellites were tested. I detected polymorphic product in 63 primer pairs, 60 of which were originally designed for birds of the order Ciconiiformes, 1 polymorphic microsatellite came from great northern loon (order Gaviiformes) and 2 from mallard (order Anseriformes). Number of alleles ranged from 2 to 8.
