Fytoplazmy jsou prokaryotní jednobuněčné organismy, postrádající buněčnou stěnu. Přežívají a rozmnožují se ve floému rostlin nebo v hemolymfě hmyzích přenašečů. V hostitelských rostlinách mohou fytoplazmy způsobit širokou škálu příznaků, jako například sterilitu, chlorózy, odumírání rostlin, zakrslost, žloutnutí nebo svinování listů, což vede ke snížení kvality a množství výnosů. Fytoplazmy jsou detekovány pomocí různých laboratorních metod, z nichž nejpoužívanější jsou metody ´nested´ PCR a kvantitativní PCR. Významným zástupcem fytoplazem je fytoplazma stolburu, která infikuje především révu vinnou a kulturní plodiny z čeledi lilkovitých a miříkovitých, a jejímž hlavním přenašečem je Hyalesthes obsoletus. Velký význam pro šíření fytoplazmy stolburu mají rezervoárové plevelné rostliny, jako například kopřiva dvoudomá a svlačec rolní. Tato diplomové práce překládá literární přehled o fytoplazmě stolburu a její genetické variabilitě. Experimentální část je zaměřena na studium genetické variability populací fytoplazmy stolburu v rostliných hostitelích (pcháč rolní, rajče jedlé, svlačec rolní, réva vinná) a hmyzím vektoru (žilnatka vironosná) pomocí amplifikace genů stamp a vmp1, sekvencování a bioinformatické analýzy. In silico RFLP analýza vmp1 genu prokázala přítomnost dvou odlišných genotypů. U izolátů fytoplazmy stolburu původem z révy vinné, pcháče rolního a Hyalesthes obsoletus byl identifikován vmp1 profil I a izolátů fytoplazmy stolburu původem ze svlačce roního byl identifikován vmp1 profil V. V rámci vybraného souboru vzorků vykazoval stamp gen vyšší genetickou variabilitu než vmp1 gen, žádný z těchto genů ale neumožnil rozlišení izolátů na základě hostitelské specifity nebo geografickém původu. Na základě analýzy haplotypů, jak v rámci stamp, tak v rámci vmp1 genu, bylo zjištěno, že populace fytoplazmy stolburu v jednotlivých hostitelích není homogenní, byla detekována různá míra jednonukletidového polymorfismu s vyšší mírou nesynonymních mutací. Populace fytoplazmy stolburu z rajčete jedlého, révy vinné a Hyalesthes obsoletus vykazovaly malou variabilitu, která vznikla v důsledku jednonukleotidových mutací pouze v rámci daného rostlinného hostitele nebo vektora. A však populace fytoplazmy stolburu z pcháče rolního a svlačce rolního vykazovaly velkou variabilitu, ke které pravděpodobně došlo směsnou infekcí.Phytoplasmas are the prokaryotic unicellular organisms lacking the cell wall. They are living parasitically in the plant phloem or in the hemolymph of insect vectors. Phytoplasmas induce in the host plants the wide range of symptoms such as steriltiy, chlorosis, necrosis and plant dying, dwarfing, yellowing or leaf roling, that could lead to the decreasing of the quality and crop yield. Phytoplasmas are detected using the various laboratory methods, the most often used methods are the nested PCR and quantitative PCR. The stolburu phytoplasma belongs to the economically important phytoplasmas. I tis infecting grapevine and various solanaceous and apiaceous cultural crops, The most important stolburu vector is Hyalesthes obsoletus Signoret. The reservoir weed plants, such as nettle and bindweed, have the great importance for the stolburu phytoplasma spreading. The theoretical part of this diploma thesis is reviewing problematics of stolbur phytoplasma and its genetic variability. The experimental part is focused on the study of genetic variability of stolbur phytoplasma populations infecting the different plant hosts (creeping thistle, tomato, bindweed, grapevine) and the insect vector (Hyalesthes obsoletus). The presence of the two different genotypes was detected using the in silico RFLP analysis of vmp1 gene. The vmp1 profile I was identified among the stolburu phytoplasma isolates obtained from the grapevine, creeping thistle and Hyalesthes obsoletus), the vmp1 profile was identified among the isolates obtained from the bindweed. The stamp gene showed in the selected set of samples the higher variability than vmp1 gene. None of them allowed the isolates discrimination according the host specificity or geographical origin. Haplotype analysis based on the both stamp and vmp1 genes showed that the stolbur phytoplasma population in the hosts is not homogeneous, the various range of single polymorphism and the higher frequency of non-synonymous mutations was detected. It was found the small phytoplasma variability of tomato, grapevine and Hyalesthes populations, this polymorphism is the result of the single mutation accumulation due to phytoplasmas replication within the host plant or vector. The higher variability was identified among the stolbur phytoplasmas identified in the populations from the creepy thistle and bindweed. The detected haplotype variability could be explained as a result of the mixed infection originating from the repetitive infection of the host plants.
