Superoxiddizmutázy sú enzýmy katalyzujúce premenu superoxidového radikálu na peroxid vodíka a kyslík. Železitá superoxiddizmutáza Fe-SOD sa u Arabidopsis thaliana vyskytuje v troch izoformách. Izoforma FSD1 doposiaľ nie je dostatočne charakterizovaná a jej úloha nie je ešte úplne známa. S využitím herbicídu paraquatu a antibiotika antimycínu A sme pozorovali, že FSD1 môže zohrávať špecifické úlohy v obrane proti oxidatívnemu stresu. S použitím antimycínu A sme zistili, že chýbajúca FSD1 u fsd1 mutantov nemá vplyv na tvorbu laterálnych koreňov a pravdepodobne nie je ani zapojená do mitochondriálnej retrográdnej signalizácie. Výrazné zníženie aktivity v koreni ale napovedá o špecifickej, no neznámej úlohe FSD1. Špecifickou indukciou oxidatívneho stresu v plastidoch s využitím paraquatu sme pozorovali zmeny v abundancii a aktivite fúzneho enzýmu GFP-FSD1 v porovnaní s FSD1-GFP a FSD1 u divého typu. S využitím konfokálnej mikroskopie sme pozorovali zníženie signálu FSD1-GFP v jadre, pravdepodobne ako dôsledok relokalizácie tohto enzýmu do plastidov a jeho následného odbúrania. Taktiež bola pozorovaná zvýšená citlivosť na paraquat u línie fsd1-1 stabilne exprimujúcej GFP-FSD1. Potvrdili sme tak dôležitú úlohu FSD1 v obrane proti oxidatívnemu stresu v plastidoch. Krížením a selekciou v F2 generácii sme pripravili rastliny fsd1-1 stabilne exprimujúce Grx1-roGFP2, ktoré v budúcnosti môžu napomôcť v štúdii FSD1.Superoxide dismutases are enzymes that catalyze the dismutation of a superoxide radical to hydrogen peroxide and oxygen. Iron superoxide dismutase Fe-SOD occurs in three isoforms in Arabidopsis thaliana. The FSD1 isoform has not yet been well characterized and its role is not fully understood so far. Using the herbicide paraquat and the antibiotic antimycin A, we observed that FSD1 may play specific roles in defense against oxidative stress. Using antimycin A, we found that the lack of FSD1 in fsd1 mutants did not affect lateral root formation and was probably not involved in mitochondrial retrograde signaling. However, a significant decrease in root activity suggests a specific but unknown role for FSD1. By specific induction of oxidative stress in plastids using paraquat, we observed changes in the abundance and activity of the GFP-FSD1 fusion enzyme compared to FSD1-GFP and FSD1 in the wild type. Using confocal microscopy, we observed a decrease in the FSD1-GFP signal in the nucleus, probably as a consequence of the relocalization of this enzyme into plastids and its subsequent degradation. Increased paraquat sensitivity was also observed in the fsd1-1 line stably expressing GFP-FSD1. We thus confirmed the important role of FSD1 in the defense against oxidative stress in plastids. By crossing and subsequent selection of the F2 generation, we prepared fsd1-1 plants stably expressing Grx1-roGFP2, which can help in the future study of FSD1.
