Tato bakalářská práce se zabývá aplikací laserové ablace ve spojení s hmotnostní spektrometrií s indukčně vázaným plazmatem (LA-ICP-MS) pro stanovení prvkových nečistot ve farmaceutických produktech. V teoretické části jsou popsány nově upravené kapitoly amerického lékopisu "232 Elemental Impurities - Limits" a "233 Elemental Impurities - Procedures", princip a instrumentace hmotnostní spektrometrie s indukčně vázaným plazmatem a laserové ablace. Součástí je rešerše shrnující využití různých technik atomové spektrometrie pro kontrolu kvality farmaceutické výroby. Experimentální část uvádí vývoj a validaci LA-ICP-MS metody pro stanovení prvkových nečistot (Cd, Pb, As, Hg, Co, V, Ni, Th, Au, Pd, Ir, Os, Rh, Ru, Se, Ag, Pt) v orálně podávaných farmaceutických produktech s doporučenou denní dávkou nepřesahující 2,0 g. Validace metody zahrnovala následující validační parametry: linearita, mez stanovitelnosti, správnost, přesnost, specificita, robustnost a rozsah. Následně byla validovaná metoda úspěšně aplikována pro stanovení prvkových nečistot v 9 účinných farmaceutických substancí, 3 vstupních surovinách a 2 finálních farmaceutické produktech.The objective of this bachelor thesis is application of laser ablation in connection with inductively coupled plasma mass spectrometry (LA-ICP-MS) for the monitoring of elemental impurities in pharmaceutical products. In the theoretical part the newly edited capitols of American pharmacopeia "232 Elemental Impurities - Limits" and "233 Elemental Impurities - Procedures" are discussed in theoretical part together with the principle and the instrumentation of inductively coupled plasma mass spectrometry and laser ablation. The last chapter of the theoretical part covers the utilization of atomic spectrometry techniques in the quality control of pharmaceutical production. The experimental part describes the development and the validation of LA-ICP-MS method for the determination of elemental impurities (Cd, Pb, As, Hg, Co, V, Ni, Th, Au, Pd, Ir, Os, Rh, Ru, Se, Ag, Pt) in orally administered drug products with a daily intake not more than 2.0 g. The LA-ICP-MS method was validated in terms of linearity, limit of quantification, accuracy, precision, specificity, robustness, and range. Finally, the LA ICP-MS method was successfully applied on the elemental impurities screening in 9 active pharmaceutical substances, 3 raw materials, and 2 final pharmaceutical products.
