Automatické zobrazovacie cytogenetické systémy využívajúce fluorescenčnú in situ hybridizáciu sa za posledné desaťročie značne zdokonalili a dostávajú sa do výskumných ale aj klinických cytogenetických laboratórií. Ich použitie pomáha zrýchliť prácu a zvýšiť presnosť odpočtu minimalizovaním ľudského faktoru. V experimentálnej časti tejto bakalárskej práce bola vykonaná optimalizácia metódy fluorescenčnej in situ hybridizácie na použitie pri automatizovanej analýze amplifikácie génu HER2 systémom Metafer scanning and imaging platform (MetaSystems). Optimalizované bolo aj nastavenie klasifikačných algoritmov, ktoré systém využíva na vyhľadávanie buniek vo vzorke. Následne za použitia optimalizovaných parametrov vzoriek a predúpravy bolo vykonané vyšetrenie na dvadsiatich pacientskych vzorkách za použitia automatického zobrazovacieho systému a porovnanie výsledkov s manuálnou metódou. Optimalizáciou bolo zistené, že najlepšia hrúbka rezu vzorky použitej na vyšetrenie je 3 m. Z reagencií používaných pri príprave vzorky bol vybraný ako najvhodnejší komerčne dostupný hybridizačný pufer a komerčne dostupný roztok DAPI. Ako ideálna voľba sondy sa ukázala komerčne dostupná sonda ERBB2(17q12)/SE 17 od firmy Kreatech alebo kit na prípravu sondy od firmy Invitrogen. Bolo vykonané porovnanie dostupných klasifikačných algoritmov a spomedzi nich bol na základe parametrov zvolený klasifikačný algoritmus HER2 TissueFISH V6B. Po vyšetrení 20 vzoriek optimalizovanou metódou a porovnaním s manuálnou sa zistilo že v 5 prípadoch sa výsledky metód líšia. Optimalizáciu sa podarilo štandardizovať metódu fluorescenčnej in situ hybridizácie a nastavenia systému pre automatickú analýzu systémom Metafer scanning and imaging platform. Z porovnania automatickej a manuálnej metódy nebolo možné vyvodiť štatistické závery, kvôli nedostatočnému množstvu použitých vzoriek. Táto práca môže slúžiť na potencionálne zavedenie automatického zobrazovacieho systému v klinickej praxi.Automatic cytogenetic imaging systems using fluorescent in situ hybridization have greatly improved in last decade and are being introduced into both research and clinical cytogenetic laboratories. Their use helps to speed up the work rate and increase the accuracy of enumeration by minimizing the human error. In the experimental part of this bachelor thesis, optimization of the fluorescence in situ hybridization method was performed for use with the automated analysis of HER2 gene amplification by Metafer scanning and imaging platform (MetaSystems). The settings of classification algorithms that the system uses to look for cells in the sample were also optimized. Subsequently, using optimized sample parameters and pre-treatment, an examination of 20 patient samples was performed using an automated imaging system and results were compared to a manual method. By optimization it was found that the best section thickness of the sample used for the examination was 3 m. From the reagents used in the preparation of samples, the most preferred commercially available hybridization buffer and a commercially available DAPI solution were selected. The ideal probe was the commercially available Kreatech ERBB2 (17q12) / SE 17 probe or Invitrogen probe kit. A comparison of the available classification algorithms was performed and the HER2-TissueFISH-V6B algorithm was chosen. After examining twenty patient samples by an optimized method and comparing with the manual, it was found that in 5 cases the test results were different. Optimization has been successful for standardization of the method of fluorescence in situ hybridization and system settings for automatic analysis by the Metafer scanning and imaging platform. From the comparison of the automatic and manual method, statistical conclusions could not be drawn due to the insufficient number of samples used. This work can be used for the potential introduction of automatic imaging system in clinical practice.
