Tato práce shrnuje poznatky o meziproduktech PDNS, možnostech jejich identifikace a diagnostiky případného enzymového defektu. Jsou zde popsány syntézy meziproduktů PDNS v ribosidické formě, záznamy MS spekter pro jejich identifikaci. Spektrální vlastnosti slouží pro diagnostiku metodami, které využívají UV detekci (CE,HPLC, spektrometrie). Pro účely analýzy metodou CE tato práce popisuje jejich elektroforetické vlastnosti. Poznatky z popsaných enzymových defektů byly využity pro vývoj screeningové metody TLC, dále pak byly popsány možnosti diagnostiky případných enzymových defektů analýzou erythrocytů.This thesis summarizes the facts about intermediates of the PDNS, the possibilities of their identification and diagnosis of appropriate enzyme defect. There are described synthesis of PDNS intermediates in the ribosidic form, the reported mass spectra are useful for the identification. Reported mass spectrometric and ultraviolet spectral properties are useful for diagnosing of patients using validated methods based on electrophoretic and liquid chromatography principles. A simple thin layer chromatographic method useful in mass screening is also reported here. These screening results can be further studied by the reported method for erythrocyte analysis.
