Na malém nížinném toku Sitka byly sledovány procesy ovlivňující koloběh metanu. Byl zjišťován metanotrofní a metanogenní potenciál vzorků sedimentu pro odlišné velikostní frakce a hloubky sedimentu. Dále byly měřeny koncentrace rozpuštěného kyslíku a metanu v intersticiální vodě ve vertikálním profilu sedimentu, difuzní toky metanu ze sedimentu a byla provedena optimalizace doby měření emisí metanu z vodní hladiny. Pro odběry vzorků sedimentu z různých hloubek byla použita namražovací sonda (?freeze-core?). Odběry vzorků intersticiální vody ve vertikálním profilu byly provedeny za použití tzv. minipiezometru. Difuzní toky metanu ze sedimentu do povrchové vody byly zjišťovány přímým měřením pomocí bentických komor. Hodnoty metanogenního a metanotrofního potenciálu byly v jednotlivých hloubkách sedimentu rozkolísané. V závislosti na velikostních frakcích sedimentu byl zaznamenán signifikantně vyšší metanogenní potenciál u zrn $<$ 6 mm. Koncentrace metanu v intersticiální vodě se zvyšovala s rostoucí hloubkou sedimentu (r2 = 0,96; P = 0,003), zatímco koncentrace rozpuštěného kyslíku v intersticiální vodě se s rostoucí hloubkou snižovala (r2 = 0,90; P = 0,013). Pozitivní difuzní toky metanu ze sedimentu do povrchové vody naznačily, že se sediment může podílet na obohacování povrchové vody metanem. Nejvhodnější dobou pro odběr vzorků metanu při měření emisí z hladiny vodního toku se jevil úsek mezi 1. a 3. hodinou trvání inkubační doby.Processes affecting the methane cycle were monitored in the small lowland stream Sitka. The methanotrophic and methanogenic potentials of sediment samples were determined for different size fractions and sediment depths. Furthermore, concentrations of the dissolved oxygen and the methane in the interstitial water in the vertical profile of the sediment were measured, as well as the methane diffusion fluxes out of the sediment. The time optimization of measurement of the methane emission from the airwater interface was also realised. The freezing probe (?freeze-core?) was used for sediment samples from various depths. Samplings of interstitial water in the vertical profile were performed using so-called minipiezometer. The methane diffusion flux from the sediment to the surface water were determined by direct measurements using benthic chambers. Methanotrophic and methanogenic potential values fluctuated within particular sediment depths. Among size fraction, significantly higher methanogenic potential was found to be in the grains $<$ 6 mm. The methane concentration in the interstitial water increased with rising sediment depth (r2 = 0,96; P = 0,003), while the concentration of the dissolved oxygen in the interstitial water decreased with increasing sediment depth (r2 = 0,90; P = 0,013). Positive diffusion fluxes of the methane from the sediment to the surface water indicated that the sediment may contribute to the enrichment of the surface water by the methane. The period between 1st and 3rd hour of incubation time appeared to be the best time for the methane sampling during the emission measurement.
