Bakalářská práce se zabývá účinkem bromovaných derivátů indolu na aktivitu aryl uhlovodíkového receptoru (AhR) a pregnanového X receptoru (PXR) v lidských buněčných liniích. Teoretická část je věnovaná jaderným receptorům, konkrétně aryl uhlovodíkovému a pregnanovému X receptoru, kde je podrobněji rozepsána jejich signalizace, ligandy a fyziologické a patofyziologické procesy spojené s těmito receptory. Další kapitola je věnována indolu a jeho halogenovaným derivátům. Praktická část se zaměřuje na studium transkripční aktivity AhR a PXR v přítomnosti vybraných bromovaných derivátů indolu. Z výsledků vyplývá, že všechny studované látky jsou schopné aktivace obou receptorů, avšak s různou účinností a potencí. Nejvyšší aktivaci AhR po 4 h působení způsobuje 6-bromindol a 7-bromoindol, zatímco po 24 h působení byl AhR nejvíce aktivován 5-bromindolem a 6-bromindolem. Nejnižší hohnoty EC50 byly pozorovány u bromoindolu v poloze 4, 6 a 7 po 4 hodinách inkubace. Co se týče PXR, nejvyšší aktivace po 24 h inkubace byla zaznamenána u 7-bromindolu, u něhož byla pozorována nejvyšší účinnost i potence.The bachelor's thesis deals with the effect of brominated indole derivatives on the activity of aryl hydrocarbon receptor (AhR) and pregnane X receptor (PXR) in human cell lines. The theoretical part delves into nuclear receptors, specifically focusing on the aryl hydrocarbon receptor and the pregnane X receptor, where their signaling, ligands and physiological and pathophysiological processes associated with these receptors are discussed in detail. The next chapter is devoted to indole and its halogenated derivatives. The practical part of this thesis focuses on the study of the transcriptional activity of AhR and PXR in the presence of selected brominated indole derivatives. The results show that all the studied substances are capable of activating both receptors, but with different degrees of efficiency and potency. The highest activation of AhR after 4 h of treatment was caused by 6-bromindole and 7-bromindole, whereas after 24 h 5-bromindole and 6-bromindole exhibited the highest activation of AhR. The lowest EC50 values were observed for bromoindole at positions 4, 6 and 7 after 4 hours of incubation. Regarding PXR, the highest activation after 24 h of incubation was observed for 7-bromindole, for which the highest efficiency and potency were observed.
