K experimentům v podmínkách in vitro byl využit genetický přístup spočívající v analýze růstových reakcí mutantů. Aquaporins, the channel proteins, facilitating the osmotic transport of water and other molecules, are important for plant growth and development. Little is known about their response to light, or whether the light causing photomorphogenesis regulates the activity of aquaporins. One way to find out was to grow plants in the presence of an aquaporin inhibitor, mercuric chloride (HgCl2) and under various light conditions. To the experiments undertaken in conditions in vitro, the genetic approach based on the analysis of growth responses of mutants was applied. Specifically, the recessive tomato mutants auW (aurea) and yg2 (yellow-green-2), both deficient in phytochrome chromophore, were used for experiments. The results of the experiments showed that blue and red light affect the sensitivity of tomato hypocotyls to the aquaporin inhibitor through the phytochrome chromophore. The results also lead to the conclusion that under all tested light conditions aquaporins sensitive to mercuric chloride are positively involved in hypocotyl and root growth of tomato plants.
