Teratogeny jsou vnější faktory, které indukují vznik vrozené vady. Za teratogeny jsou považovány léčiva, drogy, chemikálie, různé druhy ionizujícího záření a také infekční choroby. Výrazný zájem o obor teratologie a zkoumaní teratogenů vyvolala thalidomidová aféra, s jejichž následkem, vznikem vrozených vad, se potýkalo více než 10 000 dětí. Cílem rešeršní práce je z dostupných odborných článků definovat možné molekulární mechanismy teratogeneze konkrétních teratogenů. Práce se zabývá hlavně teratogenezí vybraných léčiv, včetně zmíněného thalidomidu, a také molekulárními mechanismy vzniku poruch fetálního alkoholového spektra. Thalidomid je z hlediska molekulárních mechanismů teratogeneze nejlépe prozkoumanou látkou, kdy se podařilo identifikovat i jeho pravděpodobný molekulární cíl - cereblon. Vybrané teratogeny spojuje z hlediska mechanismů teratogeneze hypotéza oxidačního stresu a změna exprese určitých genů, jež vedou k narušení vývoje tkání/orgánů a vzniku malformací.Teratogens are external factors that induce emergence of birth defects. Drugs, addictive substances, chemicals, various types of ionizing radiation and also infectious diseases are considered as teratogens. The thalidomide affair, which resulted in birth more than 10,000 children with birth defects, has aroused significant interest in the field of teratology and the study of teratogens. The aim of this review is to define possible molecular mechanisms of teratogenesis of specific teratogens from available scientific articles. The work deals mainly with the teratogenesis of selected drugs, including the aforementioned thalidomide, as well as the molecular mechanisms of fetal alcohol spectrum disorders. Thalidomide is the best examined substance in terms of molecular mechanisms of teratogenesis, even its probable molecular target - cereblon, has been identified. In terms of the mechanisms of teratogenesis, selected teratogens are connected by the hypothesis of oxidative stress and changes in the expression of certain genes, which lead to disruption of tissue / organ development and malformations.
