E3 ubiquitin ligáza RNF168 je jedním z klíčových proteinů regulujících prostřednictvím ubiquitinové signalizace opravu dvouvláknových zlomů DNA. Navzdory její nepostradatelné úloze při akumulaci efektorů jako 53BP1 a BRCA1, nebyly doposud příliš zkoumány její postranslační modifikace a jejich úloha při modulaci biologických funkcí. Cílem práce je identifikovat tyto modifikace a popsat jejich vliv na aktivitu a stabilitu proteinu RNF168.RNF168 E3 ubiquitin ligase is one of the key proteins mediating the ubiquitin-dependent signalization in the DNA double-strand break repair. Despite its indispensable role in the recruitment of downstream effectors such as 53BP1 and BRCA1 to the site of damage, post-translational modifications and their biological functions have not been investigated on detail, yet. Aim of this work is to identify these modifications and describe their role in RNF168 activity and stability modulation.
