Tato diplomová práce se zaměřuje na analýzu a charakteristiku polymorfních mikrosatelitů u plameňáka růžového. Teoretická část práce se věnuje popisu plameňáka růžového (Phoenicopterus roseus) a jeho zařazení do systému, dále se zabývá obecnou charakteristikou mikrosatelitů, jejich vlastnostmi, strukturou, mutacemi a využitím. Na závěr teoretické části jsou popsány polymorfní mikrosatelity dosud nalezené u plameňáka růžového a také nově nalezené mikrosatelity vhodné k otestování u tohoto druhu. V praktické části byla provedena PCR amplifikace vybraných mikrosatelitových lokusů s využitím primerů izolovaných jednak přímo od plameňáka růžového a jednak od příbuzných druhů z řádů brodiví (Ciconiiformes) a veslonozí (Pelecaniformes). PCR produkty byly podrobeny elektroforetické separaci v 6% polyakrylamidovém gelu a vzniklý elektroforetogram vyhodnocen. Zjištěné výsledky byly statisticky zpracovány pomocí programů Cervus 3.0.3 a Genepop 4.2. Celkem bylo charakterizováno 47 mikrosatelitů s počty alel pohybujícími se od 2 do 24. Průměrný počet alel na lokus byl 5,9. Čtyři z těchto mikrosatelitů vykazovaly vazbu na pohlaví.This master thesis focuses on the analysis and characterization of polymorphic microsatellites in Greater Flamingo. The theoretical part deals with the description of a Greater Flamingo (Phoenicopterus roseus) and its taxonomy, discusses the general characteristics of microsatellites, their properties, structure, mutations and applications. At the end of the theoretical part the polymorphic microsatellites have been found in Greater Flamingo and newfound microsatellites suitable for testing in this species are described. In the practical part of this thesis the PCR amplification of specific microsatellite loci was performed using primers isolated directly from Greater Flamingo and also from related species belonging to orders Ciconiiformes and Pelecaniformes. PCR products were electrophoretically separated in 6% polyacrylamide gel and the resulting elektroforetogram was evaluated. The results were statistically processed using Cervus 3.0.3 and Genepop 4.2. Totally, 47 of polymorphic microsatellites were characterized with numbers of alleles moving in range from 2 to 24. The average number of alelles per locus was 5.9. Four of those microsatellites were sex-linked.
