Mechanická stimulace mechanosenzitivních výběžků uvnitř pasti mucholapky podivné (Dionaea muscipula Ellis) vyvolává elektrické signály. Opakované mechanické podněty a tvorba akčních potenciálů vede k akumulaci fytohormonů ze skupiny jasmonátů, což způsobí expresi genů závislých na kyselině jasmonové, které hrají zásadní roli při obraně nemasožravých rostlin po napadení býložravcem. Následuje syntéza transportních proteinů a trávicích enzymů, které jsou sekretovány do uzavřené pasti mucholapky podivné. Cílem této diplomové práce je sledovat meziorgánovou komunikaci v masožravé rostlině mucholapce podivné, a to konkrétně šíření elektrických signálů a jejich vliv na akumulaci jasmonátů. Jakož i následnou expresi trávicích enzymů v různých typech pletiv (past vs. asimilační list) po poranění jehlou nebo mechanické stimulaci mechanosenzitivních výběžků. Pro tyto účely bylo provedeno povrchové měření elektrických signálů pomocí Ag/AgCl elektrod, kvantitativní analýza fytohormonů LC-MS/MS a plánována byla i qPCR pro analýzu exprese genů v jednotlivých částech masožravé rostliny mucholapky podivné. Výsledky naznačují, že se akční potenciál mezi pastí a asimilační částí listu nepřenáší a podle kvantitativní analýzy kyseliny jasmonové dochází k aktivaci její signální dráhy výhradně ve stimulované části rostliny. Jediným fytohormonem, který byl akumulován po poškození asimilační části listu i v pasti mucholapky podivné je kyselina abscisová. I přesto, že meziorgánová komunikace je k přežití nemasožravých rostlin esenciální, výsledky této diplomové práce naznačují, že pro mucholapku podivnou je meziorgánová komunikace postradatelná, i když ne zcela postradatelná.The mechanical stimulation of mechanosensitive trigger hairs inside the trap of the Venus flytrap (Dionaea muscipula Ellis) elicits an electrical signal. If the mechanical stimulation and the creation of action potentials are repeated, it leads to the accumulation of phytohormones from the group of jasmonates, which leads to the expression of genes that play a crucial role in response to herbivory attack in non-carnivorous plants. This is followed by the synthesis of digestive enzymes and proteins for the transport of nutrients, which are secreted into the closed trap of the Venus flytrap. This diploma thesis aims to monitor the inter-organ communication in the carnivorous plant of the Venus flytrap, specifically the propagation of electrical signals and their effect on the accumulation of jasmonates. As well as the subsequent expression of digestive enzymes in various types of tissues (trap vs. assimilation leaf) after needle injury or mechanical stimulation of mechanosensitive trigger hair. For these purposes, surface measurement of electrical signals using Ag/AgCl electrodes, quantitative analysis of phytohormones by LC-MS / MS, and qPCR were planned for the analysis of gene expression in individual parts of the carnivorous plant Venus flytrap. The results indicate that the action potential between the trap and the assimilation part of the leaf is not transmitted and according to the analysis of the jasmonic acid, the signaling pathway is activated exclusively in the stimulated part of the plant. The only phytohormone that was accumulated in the nondamaged part of the Venus flytrap was abscisic acid. Although inter-organ communication is essential for the survival of non-carnivorous plants, the results of this thesis suggest that inter-organ communication is indispensable for the Venus flytrap, although not completely.
